Background: lung cancer is the leading cause of cancer death,and lung squamous cell carcinoma(SqCLC)is a common type of non-small cell lung cancer,accounting for about 30%.Although some progress has been made in the study of lung squamous cell carcinoma,the underlying molecular mechanism of its carcinogenesis remains unclear.Therefore,it is very important to enhance the study on the mechanism of lung squamous cell carcinoma.Mi RNAs play a key role in the regulation of cancer progression and metastasis,including lung cancer.Relative stability,small size,and significant regulation of gene expression are the unique characteristics of miRNAs,which distinguish them from m RNA and become new biomarkers and potential therapeutic targets in the future.Biocredit analysis showed that the survival prognosis of patients with high miR-556-3p expression was significantly better than that of patients with low miR-556-3p expression.Considering that there are few studies on miR-556-3p at present,this study explores the effects of miR-556-3p on the invasion and migration of SQCLC cells and its potential mechanisms.Materials and Methods: First,CCK-8,scratch test and Transwell were used to detect the effects of miR-556-3p transfection on the proliferation,invasion and migration of lung squamous cell carcinoma cells(NCI-h226 and SK-MES-1),respectively.Second,bioinformatics analysis and dual luciferase reporter gene analysis were used to verify the relationship between miR-556-3p and NUAK1.Third,the regulatory effect of interacting miR-556-3p and NUAK1 on the progression of SQCLC was determined.Fourth,the role of miR-556-3p in tumor development of SQCLC was verified by HE and immunohistochemical experiments in vivo.RESULTS: It was found that miR-556-3p inhibited the proliferation,invasion and migration of lung squamous cell carcinoma cells.Mechanically,NUAK1 is the target gene of miR-556-3p,and the co-transfection of miR-556-3p and NUAK1 promotes the proliferation,invasion and migration of lung squamous cell carcinoma cells.In vivo animal and cell experiments,miR-556-3p was found to inhibit EMT.CONCLUSIONS: In this study,we found that miR-556-3p inhibited EMT by targeting NUAK1,thereby inhibiting the proliferation,invasion and migration of lung squamous cell carcinoma cells.miR-556-3p may be a prognostic biomarker and therapeutic target of lung squamous cell carcinoma.
