E-cadherin(E-Cadherin,E-Cad)and estrogen receptor(ER)in normal early pregnancy and bad pregnancy(embryonic termination,hydatidiform mole,invasive hydatidiform mole)were detected by immunohistochemical technique,and the correlation between them and pregnancy status was analyzed to explore the possible role of E-Cad and ER expression in the pathogenesis of bad pregnancy(embryonic termination,hydatidiform mole,invasive hydatidiform mole).Thus early prediction of adverse pregnancy outcome and trophoblast malignant tendency to provide a theoretical basis for endocrine therapy.Methods 52 cases of embryo termination(villus tissue),30 cases of hydatidiform mole(pregnancy tissue)and 20 cases of invasive hydatidiform mole(focus tissue)were collected from December 1,2018 to October 30,2020 in the affiliated Hospital of Qinghai University.In the same period,30 normal early pregnancy women(villus tissue)who asked for termination of pregnancy in the outpatient clinic of family planning department served as the control group.After curettage,chorionic villiand hydatidiform mole pregnancy tissues were collected,and invasive hydatidiform mole tissues were selected for operation.Based on immunohistochemical method,chorionic villi,pregnancy tissue and invasive hydatidiform mole tissues were fully collected and sliced.All sections were stained with E-Cad and ER,The score was recorded in the case.The software of SPSS24.0 and Excel was used to deal with the data in the experiment.The paper analyzes the difference between the expression of E-Cad and ER in the following conditions: normal early pregnancy villi,embryo termination villi,hydatidiform mole pregnancy tissues and invasive hydatidiform mole lesions,and the correlation between E-Cad and ER and different pregnancy status,and to explore the possible role of E-Cad and ER expression in the pathogenesis of bad pregnancy(embryonic termination,hydatidiform mole,invasive hydatidiform mole).Results 1.In four groups,normal early pregnancy group,embryo termination group,hydatidiform mole group and invasive hydatidiform mole group,the positive expression rates of E-Cad are 83.33%(25/30),96.15%(50/52),60.00%(18/30)and 25.00%(5/20).There are significant differences among four groups under the expression of E-Cad(χ~2 = 60.27,P<0.01).(1)Comparing with positive expression rate of E-Cad in normal early pregnancy group,the positive expression rate of E-Cad in the embryo termination group is relatively higher.The characteristic feature in embryo termination group and the normal early pregnancy group is that the strong positive(+)expression is very high.(2)Comparing with positive expression rate of E-Cad in normal early pregnancy group,the positive expression rate of E-Cad in the hydatidiform mole group and invasive hydatidiform mole group are significant lower(P<0.05).In the two above groups,the positive expression rate of E-Cad decreases a lot.The difference among weak positive,moderate positive and strong positive in hydatidiform mole group is small,while moderate positive(++)and weak positive(+)expression was dominant in invasive hydatidiform mole group.(3)Comparing with positive expression rate of E-Cad in hydatidiform mole group and invasive hydatidiform mole group,The positive expression rate in embryo termination group is relatively higher(P <0.05).2.The positive expression rates of ER protein in normal early pregnancy group,embryo termination group,hydatidiform mole group and invasive hydatidiform mole group were 93.33%(28/30),75.00%(39/52),56.67%(17/30)and 15.00%(3/20),respectively.The difference was significant(χ~2=60.27,P<0.01).(1)Comparing with the two groups of embryo termination group and normal early pregnancy group,the rate of embryo termination group is smaller than the other group.There is a statistically significant difference in the positive expression rate of ER between the embryo termination group and normal early pregnancy group(P<0.05).(2)The positive expression rate in normal early pregnancy group is high,and the rate in hydatidiform mole group and invasive hydatidiform mole group is relatively low and there is a significant difference in rate among there groups(P <0.05).The positive expression rate of ER protein decreases in the two groups: hydatidiform mole pregnancy tissue and invasive hydatidiform mole.In hydatidiform mole group,medium positive(++)expression mostly occupies and weak positive(+)expression is more than that of others in invasive hydatidiform mole group.(3)As for the positive expression rate of ER,there is a significant difference between the two groups of embryo termination group and hydatidiform mole group and invasive hydatidiform mole group(P<0.05).The rate in embryo termination group is higher.3.By Gamma correlation test,there was no correlation between E-Cad protein and ER protein in normal early pregnancy group(Gram 0.861,P>0.05)and embryo arrest group(Gram 0.4727,P>0.05).4、By Gamma correlation test,there was a positive correlation between E-Cad protein and ER protein in hydatidiform mole and invasive hydatidiform mole.Conclusion 1.The expression of E-Cad protein in the villi of aborted embryos is higher than that in normal early pregnancy,suggesting that the abnormal expression of E-Cad protein may affect the adhesion and differentiation of trophoblasts,reduce the normal invasiveness of trophoblasts,make the embryo superficial implantation and lead to embryo termination.The rate in normal early pregnancy is greater than that of normal early pregnancy,but there is no correlation between E-Cad and ER in the group of normal early pregnancy and embryo termination.2.Comparing with the rate in the normal early pregnancy,the expression of E-Cad and ER in hydatidiform mole pregnancy tissue and invasive hydatidiform mole focus tissue is lower.With the enhancement of trophoblast invasion,the expression intensity of E-Cad protein decreased gradually,and the expression of E-Cad protein and ER protein was positively correlated in hydatidiform mole group and invasive hydatidiform mole group,suggesting that E-Cad protein and ER protein can be used as indicators for monitoring trophoblast malignant transformation.If the detection indicates that both E-Cad protein and ER protein are
