Ketone body metabolism is an important alternative metabolic pathway for cancer cells.OXCT1 plays a significant role in tumor metabolism as a rate-limiting enzyme of ketone body utilization.However,the role and function of OXCT1 in cancer are still unclear.OXCT1 has been reported that it can promote the proliferation and migration of breast cancer with unknown mechanism.Here,we showed that OXCT1 had higher enzyme activity in breast cancer cells compared with that in human normal mammary epithelial cells.Knocking down of OXCT1 inhibited the proliferation,migration and invasion of breast cancer cells.Furthermore,OXCT1 knockdown also inhibited the expression EMT-related proteins and activation of SMADs,they were the downstream of TGF-β signaling pathway.In summary,our study indicated that OXCT1 could regulate the process of EMT in breast cancer cells through the TGF-β/SMAD signaling pathway.Completion of this project has a good theoretical and practical value for exploring the regulatory network of ketone body metabolism and its function in tumor. |
