| Background and purpose:Hepatocellular carcinoma(HCC)is one of the most common malignancies,and its global burden is increasing year by year,with an estimated incidence of more than1 million cases per year soon.Genomic studies have established a molecular map of hepatocellular carcinoma,and many cytokines and signaling pathways are involved in the development of HCC,including pancreatic progenitor cell differentiation and proliferation factor(PDPF).The role of PPDPF has been poorly studied,and its main molecular function is thought to be a key regulator of pancreatic exocrine development,which is regulated by retinoic acid and pancreatic specific transcription factor 1A.The results of EST showed that the PPDPF homologous gene was detected at high level in pancreatic cancer,liver cancer,kidney cancer and so on.The purpose of this study was to investigate the expression of PPDPF in hepatocellular carcinoma and its adjacent tissues,and to analyze the relationship between the expression of PPDPF and the clinical and pathological data and prognosis of HCC,to explore the clinical value of PPDPF as a new biomarker in the treatment of hepatocellular carcinoma.Materials and Methods:The expression of PPDPF m RNA in hepatocellular carcinoma and adjacent liver tissues was analyzed in TCGA and GTEX database by GEPIA network program.Paraffin-embedded specimens and clinical data(sex,age,tumor size,tumor number and Edmondson classification)were collected from 135 patients with hepatocellular carcinoma who underwent radical surgery and were diagnosed pathologically from2008 to 2015 in the Affiliated Hospital of Jiangsu University.The expression of PPDPF m RNA in 42 pairs of HCC tissues and adjacent liver tissues was detected by real-time fluorescent quantitative polymerase chain reaction(QRT-PCR),and 4 pairs of HCC specimens with different levels of PPDPF m RNA were randomly selected,western Blot was used to detect the expression of PPDPF.Finally,the expression of PPDPF in 135 HCC tissues and 91 adjacent non-cancer tissues was detected by immunohistochemical staining to verify the previous results.At the same time,the relationship between the expression level of PPDPF in the HCC tissues and the clinical and pathological data and the correlation with the prognosis of the patients were analyzed.Result:1.Analysis of database data showed that PPDPF m RNA expression was significantly increased in HCC tissues compared with normal liver tissues(P < 0.05).2.The results of q RT-PCR,Western Blot and immunohistochemistry showed that the expression of PPDPF in HCC was higher than that in adjacent liver(P < 0.05).3.The high expression of PPDPF protein was associated with tumor size(P = 0.003),Edmondson grade(P = 0.021),recurrence(P = 0.010)and diolame complete(P =0.023).4.Survival analysis showed that the high-expression group of PPDPF protein had a poorer prognosis than the low-expression group(P = 0.043).5.Univariate and multivariate analyses showed that the high expression of PPDPF was an independent risk factor associated with decreased survival and poor prognosis in HCC patients.Conclusion:The expression of PPDPF in HCC was significantly increased,which may be involved in the development of HCC.High expression of PPDPF is associated with poor prognosis in hepatocellular carcinoma. |
