Alsophila spinulosa is the only extant woody Fern and listed as one of the national first-class endangered plants.The stem of Alsophila spinulosa can be used for medicine.It has the functions of removing rheumatism,strengthening muscles and bones,clearing away heat and relieving cough.It is often used to treat the injury caused by falls and bruises,the cough caused by lung heat,and to prevent influenza,cerebrospinal fluid,gastritis,edema,kidney deficiency and lumbago.The juice can be used externally to treat tinea.In this study,the leaves of Alsophila spinulosa were used as the research object to explore the extraction,purification and antioxidant activity in vitro.The effect of C.elegans on Alzheimer’s disease(AD)was also studied.It provides basis for utilization and development of Alsophila spinulosa.The main results are as follows:1.By the results of single factor experiments,response surface methodology was used to optimize the extraction conditions of Polyphenols from Alsophila spinulosa.The optimal extraction conditions were:Ultrasonic power 240 W,ratio of material to liquid 1:25.00g/mL,ethanol concentration 40%,extraction time 20 min,respectively.Under the optimizied conditions,the experimental Polyphenols yield was 8.79%,which was very close to the predicted yield 8.83%2.The effect of seven Macroporous resins on the adsorption rate and desorption rate of the crude extract of Alsophila spinulosa leaves polyphenols was study by the adsorbent experiment.The results showed that AB-8 resin had better purification effect based on the adsorption rate and desorption rate.The effect of sample concentration,sample loading speed,desorption ethanol volume fraction,desorption speed on the purification of AB-8resin was further analyzed.And the optimum purification conditions were obtained as follows:the concentration of the sample was 3 mg/mL,The desorption ethanol volume fraction was 70%,the Absorption flow rate was 3 mL/min,and the Desorption flow rate was 2 mL/min.The purification of polyphenols from Alsophila spinulosa leaves by Macroporous resin was improved.Under these conditions,the content of polyphenols increased from 17.34±0.05%to 47.00±0.04%.3.The purified polyphenols showed higher antioxidation activity.The IC50value of DPPH,Hydroxyl,ABTS and Anti-lipid peroxidation were 0.055 mg/mL,0.69 mg/mL,0.05mg/mL and 1.99 mg/mL respectively.4.ASP can improve the ability of anti-heat stress and anti-oxidative stress and improve the paralysis induced by temperature.,and improve Alzheimer’s disease by improving the activity of SOD and CAT and reducing the occurrence of membrane lipid peroxidation.5.ASP improves Alzheimer’s disease by up-regulating skn-1 gene expression and activating antioxidant defense system.Meanwhile,the PI3K/Akt signaling pathway was activated to alleviate Alzheimer’s disease by down-regulating pdk-1 and akt-1 gene expression levels.
