| 【Objective】Plasmacytoid dendritic cells(p DC)are important immune effector cells,which play an important role in anti-virus,autoimmunity and tumor immunity by secreting IFN-I.The purpose of this study was to investigate the infiltration characteristics of p DC in colorectal cancer and its clinical significance.【Methods】This study included a recruiment of 87 patients who underwentsurgical treatment for colorectal cancer in Ruijin hospital during the period o2017-2018,we drew tumor tissue and normal tissue from surgical removal specimen and formed it into tissue microarray,the final sample used for analysis was 76 cases.Firstly,staining was performed on a single sample by immunohistochemistry,which was used to identify tumor tissue morphology and distinguish tumor nests from tumor stroma.Then the tissue chip was detected by immunofluorescence and fluorescence scanning.The characteristics of p DC infiltration in colorectal cancer were analyzed by chi-square test.Correlation analysis was conducted to verify the regularity and correlation between each clinical index and the presence of p DC infiltration【Results】Among the 76 patients included in the study,33 were male patients and 43 were female patients,the mean age was 61.9 years.The amount of Dukes stage A,B,C was 8,31,37.The tumor growth pattern was 24 cases of uplift and 52 cases of ulcer.52 cases was examined to be well differentiation while 24 cases was diagnosed to be pooly differentiation.Immunofluorescence sections were observed and it was found that CD123+p DC was mainly infiltrated in the tumor stroma,and there was basically no infiltration in the tumor nests.In colorectal cancer patients,a major of the patients had p DC infiltration in tumor tissues and normal tissues,and the infiltration in tumor tissues was significantly higher than that in normal tissues.In some cases,there was only a small amount of p DC infiltration between tumor tissues and normal tissues,and the difference was not significant.In some cases,there was no p DC infiltration in tumor tissue or normal tissue.Through correlation analysis with clinical data,it was found that: the proportion of invasive p DC in patients of better differentiation was significantly higher than that of poorly differentiated tumors.The proportion of infiltrating p DC in colorectal cancer with protrusive growth was significantly higher than that of tumor with ulcerative growth.p DC infiltration was significantly higher in colorectal cancer tumors with lymph node metastasis than in tumors without lymph node metastasis.【Conclusion】In colorectal cancer,p DC was mainly infiltrated in tumor stroma,with no or only a little infiltration in tumor nests.Meanwhile,the infiltration in colorectal cancer tumor tissues was significantly higher than that in normal tissues.After analysis together with clinical data,it was found that p DC infiltration was less in colorectal cancer with well differentiation type and more in tumor with poor differentiation type.p DC infiltration in colorectal cancer with lymphatic metastasis was higher than that in tumors without lymphatic metastasis.The infiltration of p DC in protrusive colorectal cancer was significantly higher than that in ulcerative tumor. |
