ObjectivesThe present study detected and analyzed MTHFR gene loci and folate metabolism levels in Lanzhou Han and Tibitan populations to explore the relationship between the distribution characteristics of loci C677 T and A1298 C of MTHFR gene,gene polymorphism and recurrent abortion in different regions and races.The results will be helpful to guide clinicians to intervene early the group of recurrent abortion by implementing individualized folate supplement schedules.MethodsIn this study,200 selected samples were divided into four groups based on region,race,and history of recurrent abortion: A1: urban group of Han Nationality without recurrent abortion,A2: urban group of Han Nationality with recurrent abortion,B1:highland group of Tibetan Nationality without recurrent abortion,B2: highland group of Tibetan Nationality with recurrent abortion.The chemiluminescencewas determined to measure serum folate levels,and t test for analyze the results.The PCR monitoring technique was used to screen the distribution of loci C677 T and A1298 C of MTHFR genes,thereby judging the level of the folate metabolism.Then,the SPSS22.0 software was used for statistical analysis,while Logist is used for correlation analysis.Results1.In terms of metabolism of serum folate levels,the means of A1,A2,B1 and B2 groups were 38.04±12.23nmol/L,30.04±15.49nmol/L,27.43±10.28nmol/L and21.48±8.9nmol/L,respectively.There is significant differences among the four groups(F=31.559,P<0.001),the A2 group was significantly lower than the A1 group,while the B2 group was significantly lower than the B1 group(P<0.05).There was no statistical difference between A1 group and B1 group,A2 group and B2 group.2.As for the polymorphic distribution of C677 T and A1298 C loci,there was a statistical difference between A1 group and A2 group,as well as between B1 group and B2 group.There was no statistical difference between A1 group and B1 group,and between A2 group and B2 group.For the allele frequencies of MTHFR gene of C677 T,there is statistically difference between A1 group and A2 group,between B1 group and B2 group(P<0.05),while there is no statistically difference between A1 group and B2 group,between A2 group and B2 group(P>0.05).3.For the allele frequencies of MTHFR gene of A1298 C,there is statistically difference between B1 group and A1 group,between A1 group and B1 group(P <0.05),while there is no statistically difference between A1 group and A2 group,between A2 group and B2 group(P>0.05).The co-mutation distribution of C677 T and A1298 C of MTHFR gene in each group are correlated with folate metabolism level(P=0.014).4.The risk grade of folate metabolism ability of the four groups showed there is significant differences of folate metabolism ability between A1 and A2,B1 and B2,A2 and B2,A1 and B2 and B1 and A2(P < 0.05),but the difference of folate metabolism ability between A1 and B1 was not statistically significant(P >0.05).5.Analysis of the difference of serum folate level among the four groups shows there is significantly difference of folate metabolism ability among normal,weak and weak groups(F = 124.604,P< 0.001).6.Logist regression analysis shows that there is signifcantly difference between with history of abortion and without the history of abortion(P <0.01,the patients with the history of abortion are more likely be exposed to the risk of the absence of folate metabolism.Conclusions1.MTHFR gene polymorphism was closely related to recurrent abortion.There exists positive correlation between recurrent abortion andfolate metabolism.2.There are more MTHFR genes C677 T with mutations of CT type and TT type than those with mutations of CC type in patients with recurrent abortion.The T allele is one of the susceptible factors leadingto recurrent abortion.3.There distribution of MTHFR gene A1298 C between Han nationality and Tibetan nationality are difference,while the susceptibility is low in patients with recurrent abortion.4.Among the related factors that affect the folate metabolism of patients with recurrent abortion,the most significant factor is the history of abortion,which has a weak relationship with age,occupation,diet and environment.
