Gopher acne compound model before and after treatment,further explore using large bearing gas soup specific curative effect and mechanism of treating acne and cytokines mechanism,in order to its application in clinical treatment of acne and lay solid foundation.Methods After feeding the adaptability of 62 to golden mice,randomly divided into two groups: the blank group(golden mouse 10)and model group(golden gophers 52),The skin on both sides of the back of each group was depilated(about 2cm×2cm);Establishment of a model of acne induced by external coating of 99% oleic acid solution on both sides of the back,And at the same time,on day 17,Gavage golden hamster ropiperidine hydrochloride capsule(0.5 mg/kg)twice daily,Five consecutive days.In the blank group,hamsters were kept normally,After 4 weeks,2 golden yellow hamsters were randomly selected,To observe the skin appearance and histopathological features,It is suggested that after the successful construction of the acne complex model,Again model in rat group randomly divided into 5groups,large bearing gas soup of high,middle,low dose group,model control group,positive drug group(tanshinone capsule),After the hamsters,Carry out statistical analysis.Results(1)Skin lesions in the back of the hamster.Acne compound after model establishment,eye observed in model group golden mouse back skin to become red color on both sides,obvious thickening of the skin surface,the local coarse,with a small amount of desquamation,scabby,pore is a significant increase in the blank group with uplift papule;At the same time,it can be seen that the food intake of the model group was reduced and the defecation volume was significantly reduced.In giving treatment after 21 days,compared with the model control group,it was found that the acne lesions on both sides of the back of golden hamster in each treatment group were alleviated to varying degrees,and the relief was most obvious in the high-dose group of Dachengqi Decoction.(2)Histopathological findings.In the blank group,the tissue staining was uniform and the structure of hair follicles was normal.There were a lot of inflammatory cells in the hair follicle stoma of the model group,the hair follicle stoma was enlarged obviously,the epidermis was thickened,and the difference was obvious.Compared with model group,the treatment group was detected microscopically significantly reduce inflammatory cells,as the carrier gas soup with the increase of drug concentration and acne skin to improve the situation were different,big bearing gas decoction high dose group curative effect best,the back skin tissue close to the blank group.Western medicine group and the large dose has no obvious difference in gas bearing soup,big bearing gas soup curative effect of low dose group of the worst close to the model group.(3)Comparison of serum TLR2、TLR4、IL-1α、TNF-α contents of rats in each group.By comparing the experimental data,found that in model group rats serum TLR2 and TLR4 alpha,TNF,IL-1-alpha levels higher than blank group is significant(P<0.01);In the model group,medication group of TLR2 and TLR4 in rat serum,alpha,TNF,IL-1-alpha content all have different degrees of cut,with large bearing gas decoction high dose group had the greatest reduction.Of TLR4,IL-1α,cut down the content of TNF-α level is found,tanshinone group higher bearing gas soup dose better curative effect,but for the influence of the content of TLR2,large dosage group than in gas bearing soup tanshinone group lowered more significantly(P<0.05).(4)Comparison of TLR2、TLR4、IL-1α、TNF-α mRNA expression in rat tissues.The comparison of the experimental results showed that compared with the expression of TLR2,TLR4,IL-1α,and TNF-αmRNA in the hamster tissues of the blank group,the expression of the model group increased significantly(P<0.01).The mRNA expressions of TLR2,TLR4,IL-1α and TNF-α in each treatment group were decreased compared with the model control group(P<0.05).The mRNA contents of TLR2,TLR4,IL-1α and TNF-α in positive drug tanshinone group were the lowest compared with those in medium and low dose Dachengqi decoction groups,and the results were statistically significant(P<0.05).Conclusion According to the results,Dachengqi decoction has a positive effect on acne,and clarifies the changing trend of Dachengqi decoction in serum and tissue of acne model animals.The mechanism may be related to its regulation of TLR2、TLR4、IL-1α、TNF-αcontent in serum and tissue. |
