objectiveBy detecting the expression level of(Peripheral blood mononuclear cells,PBMCs)NLRP3 signal pathway in peripheral blood mononuclear cells of patients with chronic heart failure,to explore the expression and correlation of NLRP3 signal pathway and its induced inflammatory response in different types of chronic heart failure patients.Methods:1.Patients with chronic heart failure(NYHA Ⅱ ~ Ⅳ),Ⅱ(nude 20),Ⅲ(nude 20)andⅣ(nude 20)admitted to our hospital from 2019 to 2020 were selected and 20 normal subjects were selected as the control group.2.The clinical data of the observed subjects were collected.The peripheral venous blood of all subjects was collected,and the plasma and mononuclear cells were extracted respectively.The mononuclear cells were purified by magnetic beads,and the purity was identified by flow cytometry.The m RNA and protein expression levels of NLRP3,ASC and Caspase-1 in PBMCs were detected by real-time fluorescence quantitative polymerase chain reaction(q RT-PCR)and Western blotting(Western blot),respectively.The level of interleukin-1 β(IL-1 β)in plasma was detected by enzyme-linked immunosorbent assay((ELISA)).3.The data were analyzed by Graphad prism 6 software,and the difference expressed by P <0.05 was statistically significant.Results1.Compared with the normal control group,the expression of NLRP3,ASC,Caspase-1 protein and the level of m RNA in PBMCs of patients with grade Ⅱ,Ⅲ and Ⅳwere significantly higher(P < 0.05),the expression of these indexes in patients with gradeⅢ and Ⅳ was higher than that in patients with grade Ⅱ(P < 0.05),and that in patients with grade Ⅳ was higher than that in patients with grade Ⅲ(P < 0.05).2.Compared with the normal control group,the plasma levels of IL-1 β in patients with grade Ⅱ,Ⅲ and Ⅳ were higher than those in patients with grade Ⅱ,Ⅲ and Ⅳ(P <0.05),the expression of these indexes in patients with grade Ⅲ and Ⅳ was higher than that in patients with grade Ⅲ(P < 0.05).Conclusion1.With the decrease of cardiac function,the expression of NLRP3 pathway of PBMCs in peripheral blood of patients with chronic heart failure increased significantly,suggesting that NLRP3 inflammatory body pathway is closely related to chronic heart failure.2.With the decline of cardiac function,the level of serum IL-1 β in patients with chronic heart failure gradually increased,suggesting that chronic heart failure is closely related to inflammation.3.These results suggest that NLRP3-ASC-Caspase-1 signal pathway may cause chronic inflammation in patients with heart failure through inflammatory factor IL-1 βand play an important role in the progression of chronic heart failure.
