Objective: To evaluate the effect of genistein(Gen)on blood-brain barrier(BBB)permeability and its mechanism in rats with cerebral ischemia reperfusion.Methods: 1)72 experimental male rats were sorted into sham-operation group(Sham group),Gen pretreatment and ischemia-reperfusion group(Gen group),ischemia-reperfusion group(I/R group)by the random-number table,the number of each group is 24.In the Gen group,the animals were feed Gen by gavage once a day at a dosage of 50 mg/kg,and the remaining groups were switched to the same amount of normal saline for 3 weeks of uninterrupted intervention.On the 22 nd day,the focal cerebral ischemia-reperfusion injury(CIRI)model was established by the thread-embolization method.After 120 min of ischemia and 1d for reperfusion,the samples were detected.2)The neurological-defects of the experimental animals were evaluated by longa grade-point standard.The moisture-content of brain tissue was evaluated by the wet-dry weight means.The BBB’s permeability was evaluated by the EB-staining means.The infarct-volume condition was detected by TTC-staining method.The ultrastructural status of BBB was checked by the electron-microscopy means.The levels of cytokines IL-1β and TNF-α were detected by ELISA.The levels of NF-κB,Claudin-5 and MMP-9 were measured by WB.Results: By comparison with sham group,the neurofunctional-defect score(P<0.001),the brain’s moisture content(P<0.01),EB content(P<0.001),the infarction-volume(P<0.01),the brain-ultrastructural damage degree and the level of IL-1β,TNF-α,MMP-9 and NF-κB in the I/R group were overtly increased,while the Claudin-5’s level was visibly alleviated(P<0.001).In contrast to the I/R group,the neurological-function score(P<0.01),the brain’s moisture content(P<0.05),the EB content(P<0.01),the infarction-volume(P<0.05),the degree of the brain-ultrastructural damage and the expression of IL-1β,TNF-α,MMP-9 and NF-κB were notably relieved in the Gen group,while the protein level of Claudin-5 was markedly raised(P<0.01).Conclusion: Gen could decrease the BBB’s injury and protect CIRI,which may be necessarily bound up with participating in the regulation of Claudin-5,NF-κB,MMP-9,IL-1β.and TNF-α.
