Research purposes: Attention deficit hyperactivity disorder(ADHD)is a highly hereditary neurodevelopmental disorder.At present,the etiology and pathogenesis of ADHD are not very clear,but many theories have proposed the pathogenesis of ADHD.Many researchers believe that it is the result of genetic and environmental factors,among which the imbalance of gene expression involves tiny RNA(microRNA).The purpose of this study was to study the expression level of microRNA-5692 b and microRNA-18a-5p in patients with ADHD and their prognosis after treatment,so as to explore the clinical value of these two kinds of miRNA in the clinical diagnosis of attention deficit hyperactivity disorder(ADHD).Research methods: 1.A total of 40 children with ADHD were collected from January 2019 to June 2020,aged between 6 and 14,and 60 healthy children,aged between 6 and 14,were examined in the outpatient clinic.2.The total RNA,of serum samples of children in ADHD group and healthy control group were extracted and the expression levels of microRNA-5692 b and microRNA-18a-5p in serum samples of children in two groups were detected by q RT-PCR(real-time photoluminescence PCR).3.SPSS26.0 software was used to conduct statistical analysis and comparison on the general information(age,height,weight,gender)of the two groups of children and the expression levels of these two miRNAs,to determine the differences between the two groups and conduct statistical test.The P<0.05 is statistically significant.4.The children in the ADHD group who were willing to receive treatment were treated with methylphenidate for a period of three months,and the serum samples were collected after treatment.5.RNA was extracted from the serum samples of children in ADHD group after treatment,and the changes of microRNA-5692 b and microRNA-18a-5p in serum were detected by q RT-PCR.6.The data were analyzed and screened by SPSS26.0 software,and the differences between microRNA-5692 b and microRNA-18a-5p were compared and analyzed.7.The dynamic curves of cyclic threshold(cycle threshold,Ct)and fluorescence values were automatically analyzed by computer analysis software.Calculate the Ct value of the two groups of samples,Ct =(objective miRNA Ct-internal reference Ct)is the relative quantity of each sample 2-Ct,ADHD group and healthy control group using 2-Ct to express their 2 differences,that is,2-Ct =(Ct experimental group-Ct control group).ROC curve was used to evaluate the diagnostic value of microRNA-5692 b and microRNA-18a-5p in attention deficit hyperactivity disorder.Research results: 1.Micro RNA-5692 b is differentially expressed in ADHD children and normal children,and the expression of microRNA-5692 b in serum of children is significantly higher than that of normal children.2.Micro RNA-18a-5p is differentially expressed in ADHD children and normal children,and the expression of microRNA-18a-5p in serum of children is significantly lower than that of normal children.3.After drug treatment,the expression of microRNA-5692 b and microRNA-18a-5p in serum of ADHD children showed no significant difference from that of normal children.Analysis conclusions: Based on the results of this study,microRNA-5692 b and microRNA-18a-5p may serve as potential diagnostic and therapeutic biomarkers for children with ADHD.
