| Background:Non-alcoholic fatty liver disease(NAFLD)is a highly prevalent chronic liver disease,and the age of onset tends to be younger,which is a serious threat to human health.The medical circles at home and abroad have not yet developed a safe,effective and non-toxic side-effect drug or treatment plan for the targeted treatment of NAFLD.In recent years,bioactive peptides prepared by enzymatic hydrolysis of food-derived proteins have attracted much attention.As a typical biologically active peptide,corn peptide has a prominent role in anti-oxidation and liver protection due to its unique amino acid composition,providing a possibility for intervention in NAFLD.Purpose:This article explores the improvement effect of corn peptides on non-alcoholic fatty liver disease by establishing cell models and animal models,by measuring the expression of cytokines and related proteins and some related physiological and biochemical indicators,and performing pathological section analysis,from inflammation and oxidation The pathogenesis of Oxidative Stress(OS),Insulin resistance(IR),and lipid accumulation was started,and the mechanism of corn peptide targeted improvement of NAFLD was explored in depth.Research content and results:(1)Establish an inflammatory cell model: use LPS at a concentration of 2 μg/m L to induce macrophages RAW 264.7.Prepare three different concentrations of corn peptides(10 mg/m L,20 mg/m L,30 mg/m L)respectively.After acting on the inflammation model cells,the MTT test results show that the proliferation effect of macrophages increases firstly as the concentration of corn peptide increases.When the concentration is 20 mg/m L,the effect is the best,indicating that the corn peptide in this experiment has the greatest impact on the proliferation of macrophages when the corn peptide is 20 mg/m L.Enzyme-linkedimmunosorbent assay results showed that compared with the normal control group,corn peptides significantly reduced the inflammatory factors Tumor necrosis factor-α(TNF-α)and interleukin-1(Interleukin,IL-1)in the model group,Interleukin 6(Interleukin,IL-6)expression level,and at the same time increase the expression level of the anti-inflammatory factor Interleukin 10(IL-10),the results show that corn peptide can inhibit the cytokine in the inflammatory state.Expression,thereby reducing inflammation,further inhibiting the occurrence and development of insulin resistance and oxidative stress,and improving NAFLD.(2)Establish NAFLD cell model: use 0.6m M oleic acid to induce HL-7702[L-O2]human normal hepatocytes.After pretreatment,corn peptide acted on NAFLD model cells.Western blot analysis showed that corn peptide could intervene NAFLD by reducing the expression of sterol regulatory element binding protein 1C(SREBP-1C),increasing the expression of Adenosine monophosphate-activated protein kinase(AMPK),inhibiting ACC activity,promoting fatty acid oxidation and inhibiting lipid accumulation.The NAFLD steatotic cell model also exhibited insulin resistance,which was characterized by inhibition of IRS/Akt phosphorylation in the insulin signaling pathway and significantly decreased glucose uptake.The intracellular IRS/Akt phosphorylation level was restored after zeatin intervention,which proved that zeatin can improve insulin resistance.At the same time,it can reduce the expression of NF-κB transcription factor,reduce the intracellular Reactive oxygen species(ROS)content,reduce the inflammatory response of liver cells,relieve oxidative stress,and play a role in improving NAFLD.(3)Establish NAFLD mouse model: Use long-term high-fat diet to induce ICR mice.The results showed that corn peptide could reduce the body weight and liver index of mice by intragastric test.Serum Triglyceride(TG)and Total cholesterol(TC)levels were decreased significantly in model mice.Meanwhile,the contents of Alanine aminotransferase(ALT)and Aspartate aminotransferase(AST)in serum of mice in model group were decreased.The results indicated that corn peptide could promote fat metabolism and alleviate liver injury.Through glucose tolerance test and serum insulin concentration detection,it was verified that corn peptide could effectively improve insulin sensitivity and alleviate insulin resistance in mice.Corn peptide lavage treatment can improve model of mice liver SOD(Superoxide dismutase,SOD),Glutathione peroxidase(Glutathione peroxidase,gsh-px),and can significantly reduce the Malondialdehyde,Malondialdehyde(MDA)content,show that corn peptide has the oxidation resistance,can alleviate the liver oxidative stress,further improve the liver damage.In addition,the expression of IL-1,IL-6 and TNF-α in serum of mice was decreased by corn peptides,indicating that corn peptides could inhibit the inflammatory response caused by high fat diet.The results of liver pathology showed that corn peptide improved the degeneration of liver adipocytes. |
