Prostate cancer is the most common tumor in the male reproductive system.Its incidence is second only to lung cancer in Europe,America and other countries,and ranks second in the incidence of male tumors.In recent years,with the increasing aging of the Chinese population and prolonged life expectancy,the incidence of prostate cancer has also been increasing year by year.At the same time,because of the particularity of prostate cancer,its progression in the course of the disease is not only invasion and metastasis,but is often accompanied by changes in tumor pathology.From HNPC to CRPC to NEPC,the difficulty in diagnosis and treatment gradually increases.Therefore,the early diagnosis and screening of prostate cancer are of great significance to the treatment of prostate cancer and the control of the progression of the disease.The early treatment of prostate cancer is mainly surgical resection.Whether surgical resection directly affects the prognosis of patients,the scope of resection basically depends on the clinical experience of the surgeon.Unclean tumor resection is an important factor for tumor recurrence.Molecular imaging technology uses specific probes to perform non-invasive cellular and tissue-level functional imaging using influencing methods to meet medical diagnosis of diseases,intraoperative navigation and prognostic monitoring,and it is playing an increasingly important role in medical diagnosis and treatment.The role of.Near-infrared fluorescence imaging is widely used in the diagnosis and treatment of tumors because of its convenience,specificity,sensitivity,non-invasiveness,penetrability,and a series of advantages.According to relevant literature reports,many fluorescent probes have been used in the diagnosis of tumors in the near-infrared dye family.ICG has been used for fundus angiography,liver cancer diagnosis,intraoperative navigation,and lymph node dissection since the 1990 s.Etc.;IR-783 and MHI-148 have also been shown in a number of studies that they have tumor targeting and have the potential to be used as molecular imaging diagnostic probes;DZ-1 is a new type of Qijiachuan patented synthesis in the pre-laboratory Cyanine near-infrared fluorescent dye and its application in the diagnosis of liver cancer.At present,there are few studies on the application of heptamethine cyanine near-infrared and near-infrared fluorescent dyes in the diagnosis of prostate cancer,and it is urgent to screen the near-infrared fluorescent probes specific for prostate cancer.In this study,ICG,IR-783,MHI-148 and DZ-1 four heptamethine cyanine near-infrared fluorescent dyes were selected,respectively,from the optical properties of the compound,the targeting of tumor cells in vitro,the targeting of subcutaneous tumor dyes in vivo,and the possible existence.Comparing the research on the therapeutic potential of prostate cancer,we screened out compounds with promising applications in clinical diagnosis and treatment of prostate cancer.Firstly,UV absorption spectroscopy and fluorescence spectroscopy were performed on the four compounds,and the Living imaging equipment was used to verify the fluorescence signal intensity.Secondly,the four dyes were incubated with the prostate cancer cell line(PC-3/22RV1)to observe their presence in the tumor.Cell uptake;Next,the tumor cell line was subcutaneously transplanted into nude mice,and 4 kinds of fluorescent dyes were injected respectively,and the enrichment and targeting of the dyes in the animal body were dynamically observed through Living imaging;finally,the prostate of the 4 kinds of fluorescent dyes was explored by MTT detection Cancer treatment potential.Studies have found that four compounds,ICG,IR-783,MHI-148 and DZ-1,have relatively stable optical characteristics,and have better UV absorption in organic solvents and serum;the specific adsorption of dyes shows that except for ICG,The other three dyes can bind to tumor cells,but DZ-1 has the strongest fluorescence;in the study of prostate cancer cell PC-3subcutaneous tumor-bearing tumor targeting study,the results are consistent with the cell-level results,except that ICG is not rich in tumors.Set,the other three dyes all show certain tumor targeting;in the MTT results,ICG,IR-783 and MHI-148 did not show tumor suppression effects,but high concentrations of DZ-1 showed certain tumor suppression effects.Through various studies and comparisons,DZ-1 is a near-infrared fluorescent dye that shows certain advantages over the other three compounds in the diagnosis and treatment of prostate cancer.It has shown certain advantages in the diagnosis and treatment of prostate cancer in clinical practice.The prospect of application.Tumor-specific targeting,strong fluorescence properties,and certain tumor suppression conditions make DZ-1 fluorescent dyes expected to be used as an integrated medicine in diagnosis and treatment to improve the diagnosis,treatment and surgical navigation of prostate cancer.
