Inonotus obliquus is a kind of rare medicinal fungus originated from Jilin and Tibet of China.It has many different physiological activities,and its anti-tumor activity is the most powerful.The triterpenoids in Inonotus obliquus have a very good inhibitory effect on tumor cells.However,most of today’s anti-tumor chemotherapy drugs are toxic,which is very easy to cause harm to human body and is not conducive to the treatment of tumor.It can not only reduce the toxicity,improve the anti-tumor effect,but also save the cost to extract the monomer compounds with anti-tumor activity from the natural Inonotus obliquus.In this study,the ethanol extract of Inonotus obliquus from Nyingchi Tibet was used as material to study its inhibitory effect on NSCLC.It was found that it had a good inhibitory effect on NSCLC,which could inhibit cell migration and reduce cell clone formation.However,the crude extract is still a mixture,so there are many active substances with unknown components in the crude extract.In this way,it is difficult to find the active monomer which plays a major role.Based on this,this study was carried out by ethyl acetate extraction from ethanol extract of inorhus obliquus,and separation and purification of ethyl acetate by gel column chromatography,normal silica gel column chromatography and thin layer chromatography(TLC).The compounds were further identified by NMR and other modern mass spectrometric techniques.The 10 compounds isolated and identified are the main active components.In the follow-up experiments,the preliminary screening of cell proliferation and cell migration will be carried out.It is found that HH-1 and hh-2 have anti-tumor activity.Finally,the anti lung cancer activities of HH-1 and hh-2 were evaluated by cell migration and cell proliferation.The main results are as follows:The results showed that the crude ethanol extract of P.fuscata had anti-tumor activity against A549 and SPCA-1,significantly inhibited the proliferation of A549 and SPCA-1,inhibited the migration of A549 and SPCA-1,and reduced the formation of the two kinds of cells.The crude ethanol extract was extracted,purified and identified,and10 compounds were obtained.A total of 20 monomers were isolated from Pseudomonas obfuscata,and 10 compounds were analyzed and identified as follows: Inotodiol(HH-1),ursolic acid(HH-2),betulinol(HH-3),vanillic acid(HH-4),syringic acid(HH-5),trans caffeic acid betulinol(HH-6),protocatechuic acid(HH-7),gallic acid(HH-8),osthol(HH-9),and dihydrocelinol angelic acid(HH-10),The structural formula of the other compounds is still being analyzed.The results showed that HH-1 inhibited the proliferation and migration of A549 cells at the concentration of 200 μmol/L and had anti-lung cancer activity.HH-2 can inhibit the proliferation and migration of SPCA-1 cells at a concentration of 200 μmol/L,and it also has anti-lung cancer activity.Furthermore,HH-1A549 could significantly inhibit cell proliferation and migration in a concentration-dependent manner through cell proliferation and migration experiments.HH-2 can significantly inhibit the proliferation and migration of SPCA-1,and the effect becomes better with the increase of the concentration.
