Curative Effect Of Oxycodone Sustained-release Tablets Combined With Duloxetine Versus Paroxetine In Treatment Of Depression And Pain From Advanced Cancer

Objective:To study the curative effect and safety of two kinds of antidepressants combined with oxycodone sustained-release tablets in the patients with advanced cancer pain and depression.Methods: 108 patients with advanced cancer pain depression recruited in Hubei Zhongshan Hospital from July 2018 to September 2019 were collected in this study.All patients were divided into 3 groups of 36 patients each via random and single-blind way.The groups included Group C(oxycodone sustained-release tablets),Group D(paroxetine+the tablets)and Group F(duloxetine+the tablets).In different times after treatment,Anxiety,depression,pain relief,quality of life,sleep quality,dose of oxycodone,side effects and patient satisfaction were compared between all groups.Results: The numeric rating scale(NRS)score of Group F was significantly lower than those of Group D and C after treatment of 7 days(P<0.05).The self-rating anxiety scale(SAS)score and NRS score at 14 days after treatment,and the self-rating depression scale(SDS)score at 7 days after treatment were decreased in a trend significantly from Group C to Group D to Group F(all P<0.05).However,there were no significant differences in the SDS score at 14 days after treatment between Group F and D,and the SAS,SDS and NRS scores at 1 and 2 months after treatment(P>0.05).All of these scores were significantly lower than those of Group C(P<0.05).After treatment of 2 months,the average daily oral dose of oxycodone per capita in Group C and D were higher than that before treatment significantly(P < 0.05),but no such difference was seen in Group F(P > 0.05).No significant differences were seen in the average daily oral dose of oxycodone per capita and the score of Pittsburgh sleep quality index(PSQI)score between Group F and D,and the dose and score were both lower than those of Group C significantly(P<0.05).And after treatment,the PSQI score were significantly decreased in the 3 groups(P < 0.05).The scores of different items of quality of life score were remarkably increased in the 3 groups in2 months after treatment(P<0.05),but there were no differences in other scores between Group F and D except for the role function score(P>0.05),and the scores were remarkably higher than those of Group C(P<0.05).The side effects had no significant differences between the 3 groups after treatment of 2 months(P > 0.05),and were reduced or disappeared gradually after dose adjustment or symptomatic treatment.After treatment of 2months,there were no differences in patient satisfaction between Group C and D,Group D and F(P>0.05),but those of Group F were higher than those of Group C(P<0.05).Conclusion: Duloxetine combined with oxycodone sustained-release tablets has a quicker onset than paroxetine in the treatment of depressive patients with advanced cancer pain.And it can improve anxiety,depression,pain,quality of life and sleep,and patient satisfaction effectively,reduce dose of oxycodone,and does not increase side effects.
