Objective: To explore the correlation between PD-1 polymorphism and occurrence of hepatitis B in Ningxia through the analysis of gene single nucleotide polymorphism(SNP)of programmed death receptor 1(PD-1)in hepatitis B patients and normal population in Ningxia,so as to provide new ideas and directions for the treatment and prevention of hepatitis B,and to further provide the corresponding theoretical basis for PD-1 inhibitors in the treatment of hepatitis B virus in the future.Methods: To analyze PD-1 gene polymorphism and hepatitis B susceptibility in Ningxia,healthy subjects and hepatitis B patients who visited the People’s Hospital of Ningxia Hui Autonomous Region from January 2018 to March 2020 were included.The inclusion and exclusion criteria were set per the Guidelines for the Prevention and Treatment of Chronic Hepatitis B(2015 Updated Version).The patients were divided into control group(n = 147),hepatitis B carrier group(ASC)(n = 62),liver cirrhosis group(LC)(n = 72),chronic hepatitis B group(CHB)(n = 74)and chronic acute liver failure group(ACLF)(n = 60).Peripheral EDTA anticoagulant whole blood and corresponding serological parameters were collected,together with the extraction of whole blood DNA of the experimental group and the control group.The concentration and purity of nucleic acid were then adjusted by Nano Drop.Moreover,the locus sequence was detected by SNapshot method on account of the principle of single base extension.Besides,the differences in alleles,genotypes and serum liver function between the experimental group and the control group were analyzed by statistical methods.Results: The C allele with Rs10204525 reduces the risk of HBV infection(OR=0.339,95%CI=0.251-0.459,P=0.038),The G allele of Rs2227982 is an inductive factor of HBV infection,(OR=2.375,95%CI=1.768-3.190,P=0.014).Genotype frequency analysis between experiments and controls showed that the genotype CC 、 CT of Rs10204525 is a protective factor of HBV infection,and that the genotype GG 、AG of Rs2227982 is a risk factor for HBV infection.Conclusion: The Rs10204525 and Rs2227982 of PD-1 SNP are associated with the risk of HBV infection.
