Objective To investigate the expression changes of miR-203a-3p in pathological tissues of normal esophageal squamous epithelium,intraepithelial neoplasia,esophageal squamous cell carcinoma in patients with esophageal squamous cell carcinoma,and the relationship between miR-203a-3p expression level and clinical data,inflammatory markers,tumor markers of patients with esophageal squamous cell carcinoma,to provide evidence for miR-203a-3p as a biomarker for early diagnosis and disease follow-up of esophageal squamous cell carcinoma.Methods Postoperative paraffin tissue specimens and related clinical data of 30 patients with esophageal squamous cell carcinoma clinically archived in Minda Hospital of Hubei Minzu University from January 2018 to October 2020 were collected.The expression levels of miR-203a-3p in normal esophageal squamous epithelial,intraepithelial neoplasia,esophageal squamous cancer were detected respectively by microdissection,quantitative real-time polymerase chain reaction(q RT-PCR).The correlations between the expression level of miR-203a-3p in esophageal squamous cell carcinoma and clinical datas,tumor markers,inflammatory markers of patients were compared,and its diagnostic value in esophageal squamous cell carcinoma was evaluated.Results 1.The expression level of miR-203a-3p in esophageal squamous cell carcinoma tissues was significantly lower than that in normal esophageal squamous epithelial tissues,and the difference was statistically significant(P < 0.01);The expression level of miR-203a-3p in intraepithelial neoplasia tissues was significantly lower than that in normal esophageal squamous epithelial tissues,and the difference was statistically significant(P<0.05);The expression of miR-203a-3p in 63.4%(7/11)esophageal squamous cell carcinoma tissues was lower than that in intraepithelial neoplasia tissues,but there was no statistically significant difference in the expression level of miR-203a-3p in the two tissues(P>0.05).2.The expression level of miR-203a-3p in esophageal squamous cell carcinoma tissues was negatively correlated with the degree of invasion depth(P<0.05),not significantly correlated with the age,gender,tumor size,tumor differentiation,growth site,lymph node metastasis and pathological stage of patients(P>0.05).3.Correlation analysis was conducted between the expression level of miR-203a-3p in esophageal squamous cell carcinoma tissues and inflammatory markers,tumor markers detected in peripheral blood,the result showed that the expression level of miR-203a-3p was negatively correlated with CEA(P < 0.05),not significantly correlated with AFP,CA199,CA724,SII,NLP,PLR(P> 0.05).4.The ROC curve was used to analyze the diagnostic value of miR-203a-3p in esophageal squamous cell carcinoma,and found that the AUC of the area under the ROC curve for miR-203a-3p expression was0.941(95%CI: 0.885 ~ 0.996).When the critical value was 4.98,the diagnostic value of miR-203a-3p was the highest,with sensitivity and specificity were 87.8%and 96.7%,respectively.Conclusion 1.The low expression of miR-203a-3p is helpful for the diagnosis of esophageal squamous cell carcinoma.2.The expression level of miR-203a-3p is negatively correlated with the infiltration depth of esophageal squamous cell carcinoma,which has certain reference significance for the follow-up of patients with esophageal squamous cell carcinoma. |
