| Cornus officinalis is a traditional Chinese medicinal material,which has the functions of nourishing kidney and liver,nourishing qi and blood,lowering blood sugar,anti-inflammatory,and neuroprotection.Cornus iridoid glycosides(CIG)are characteristic components of Cornus."Chinese Pharmacopoeia" regards morroniside and loganin as the main medicinal components of Cornus.This research focuses on the separation,purification,identification,and cytological evaluation of iridoid glycosides.The dried fruit of Cornus officinalis was crushed and extracted with methanol by ultrasonic extraction;the crude extract was passed through D101 macroporous resin to remove impurities such as pigments and enrich iridoid glycosides;The ion trap mass spectrometry was used to identify the main components of the extract,and then quantitative analysis was detected by high performance liquid chromatography and triple quadrupole mass spectrometry;High concentration of glucose induces apoptosis of vascular endothelial cells.The extract is added to cell culture medium at different concentrations to explore its effect on cell proliferation,morphology,and apoptosis.The results showed that,cornus officinalis was extracted by ultrasonic and enriched with macroporous resin,and freeze-dried to be a powder.The final product contained 1.6% ash,5.31% water,and 35.44%polysaccharide.The four main compounds could be determined by high-performance liquid phase and mass spectrometry.The content in the extract was determined to be 28.5% of morroniside,20.4% of loganin,4.5%of cornoside,about 2% of seriogenin,and the total content of iridoid glycosides exceeded 55%.The CIG extract was added to the cell culture system,and it was found that the appropriate concentration of CIG can significantly promote the proliferation activity of vascular endothelial cells;High concentration of glucose could significantly inhibit cell activity,leading to cell atrophy and apoptosis;CIG extract was added into this culture system,and it was found that CIG at an appropriate concentration could improve the cell atrophy and inhibit the decrease of cell proliferation activity;The early and late stages of apoptosis showed that the intervention of CIG could inhibit cell apoptosis and reduce the proportion of late apoptosis and necrosis.The combination of ultrasonic extraction and macroporous resin enrichment could obtain cornus iridoid glycolsides with higher purity.Intervention of CIG extract into cell culture can significantly improve the activity of vascular endothelial cells.The integrity of the monolayer of vascular endothelial cells is the key to vascular endothelialization,and if it was destroyed,the extracellular matrix would be exposed.The collagen and insoluble fibrin in the extracellular matrix would cause platelets to rapidly agglutinate and form thrombus.Cornel extract improves the activity of vascular endothelial cells,can prevent vascular endothelial cells from shedding and apoptosis to a certain extent,and promote vascular endothelialization.Hyperglycemia,diabetes,etc.could lead to a series of cardiovascular and cerebrovascular diseases.The reason is also related to the high sugar in the environment of vascular endothelial cells.The extract of Cornus officinalis could inhibit the apoptosis of vascular endothelial cells caused by these diseases,and could also relieve and improve a series of complications caused by high glucose. |
