Purpose: To observe the safety and effectiveness of patching retinal breaks with ocular linear hydrogel combined with 25 gauge vitrectomy and sterile air filling in the experimental rhegmatogenous retinal detachment.Methods:(1)Cohesion: The cohesion of ocular linear hydrogel and other hyaluronic acid were compared by dispersing method.The products were stained with toluidine blue and pushed into a beaker containing plasma water,and the beaker was evenly stirred to observe the dispersion of each product.(2)Safety: After 25 gauge vitrectomy and air-fluid exchange in the left eye of eight healthy adult gray rabbits,0.1ml linear hydrogel and sterile air were filled.The change of ocular condition after operation was observed.Preoperative and postoperative ERG was applied to monitor the functional variety.Retinal sections were examined by HE staining to detect the changes of retinal morphology.(3)Effectiveness: The left eyes of 16 healthy adult gray rabbits were randomly divided into experimental group and control group.After 25 gauge vitrectomy,a retinal break about 1/2 PD size was constructed below the optic disc with vitrector,subretinal injection of balanced saline solution through the break caused retinal detachment with a range of about 2 optic disc diameters.Following air-fluid exchange was carried out,after sterile air filled vitreous,patching retinal break with ocular linear hydrogel in experimental group,and the control group without the procedure.The retinal reattachment and complications of two groups were evaluated at 1 week,1 month and 3 months after operation,with the help of fundus photography and B-ultrasound.Outcome:(1)The cohesion of ocular linear hydrogel was significantly stronger than that of the other two commonly used hyaluronic acid.(2)There was no significant change in retinal morphology and function after the vitreous was filled with ocular linear hydrogel comparing with contralateral rabbit eye.Filling vitreous cavity with ocular linear hydrogel in rabbit eye is safe,without toxic and side effects.(3)During the 3 months of follow-up,there was no infected eye in experimental and control group,all of the retina reattached in 8 eyes of experimental group,there was 2 eyes with retinal reattachment and 6 eyes with retinal detachment in the control group.There was no significant complication in two groups.Conclusion: Patching retinal breaks with ocular linear hydrogel combined with 25 gauge vitrectomy and sterile air filling in the rabbit rhegmatogenous retinal detachment model is safe and effective. |
