| Objective:Metabonomics detection technique was used to observe and analyze the characteristics of metabonomics in patients with different degrees of coronary artery stenosis.To explore the difference of metabolites between the coronary heart disease group and the reference group and the difference of metabolites between the patients with different degrees of coronary artery stenosis.Methods:Patients admitted to the Department of Cardiology of Tianjin Medical University General Hospital from 12 th 2019 to 1th 2020 with chest pain as the main symptom and suspected coronary heart disease were enrolled in the study.According to the degree of coronary artery stenosis,the patients were divided into the following four groups: the stenosis <50% group was defined as the stenosis of the three main coronary arteries(including the anterior descending branch,the circumflex branch and the right coronary artery)<50%;the stenosis 50-75% was defined as at least one lesion with at least one main coronary artery was ≥50% and <75%;the stenosis75-95% was defined as at least one lesion with at least one main coronary artery ≥75% and <95%;the stenosis ≥95% was defined as at least one lesion with at least one coronary artery stenosis ≥95%.20 patients were selected in each group.In addition,patients with coronary artery stenosis <50% were defined as reference group,and the coronary heart disease group was defined as more than 50% stenosis in one lesion with at least one main coronary artery.The clinical data of all patients including age,sex,previous medical history,routine biochemical examination,electrocardiogram,echocardiography and intraoperative coronary angiography were recorded.All patients were treated with invasive therapy or conservative drug therapy according to clinical guidelines to alleviate myocardial ischemia and prevent myocardial infarction.All patients kept fasting serum samples of 8 Mel for 12 hours and stored at-80 ℃.The serum samples were detected by mass spectrometry chip in metabonomics technology,PCA and OPLS-DA were used in unsupervised and supervised pattern recognition.Pairwise analysis and comparison between the CHDgroup and the control group,and among the four groups,exploring the differences of metabolites between them.Results:The main results were as follows:(1)There were differences in lysophosphatidylcholine,cyclophosphatidic acid,tryptophan carnosine and so on between the CHD group and the reference group.(2)Comparing the four groups with different degrees of coronary artery stenosis,five different metabolites were found,including taurine,malic acid,malic acid,isovalerate,lysophosphatidic acid [LPA(8:0/0:0)] and cholesterol linoleic acid[18:2(9Z,12Z)].Conclusion:Metabonomics detection technology found that there are indeed different metabolites in patients with different degrees of coronary stenosis,and has diagnostic value.In addition,the metabolic disorders involved in coronary heart disease may be systemic,including energy metabolism and substance metabolism disorders.With the application of metabonomics technology,its pathophysiological mechanism was analyzed in order to obtain new targets and new ideas for diagnosis and treatment. |
