OBJECTIVE: To research the composition of gastric microbiota of patients with chronic gastritis,and compare the distribution of gastric microbiota of H.pylori negative and H.pylori positive,and analyze the relationship between H.pylori and the patient’s age,gender,chronic atrophic gastritis(CAG).We analyzed the etiology and pathogenesis,syndromes and progress of traditional Chinese medicine treatment of chronic gastritis patients.To provide an effective way for chronic gastritis.Methods: Collecting the clinical data of patients with chronic gastritis who were treated in the Endoscopy Center of Tianjin People’s Hospital,and signing the informed consent after met the inclusion and exclusion criteria.All Participants was performed with 14C-UBT and pathology.The gastric mucosa tissue was centrifuged to obtain a precipitate and DNA was extracted by using a kit.Amplifying the V3 / V4 region of the 16 s rRNA region of DNA with primers,and then sequencing.Analyzing the patients with chronic gastritis through tongue coating,pulse phase,clinical symptoms and other data to determine TCM syndromes and etiology and pathogenesis.Using software to analyze gastric microbiota.Data analysis was performed using SPSS 23.0 statistical software.P < 0.05 was considered statistically significant.Results: 1.The four most abundant phyla represented more than 90% of the observed sequences and were found in all samples: Firmicutes,Proteobacteria,Bacteroidetes and Actinobacteria;At the family level,family Streptococcaceae,Veillonellaceae,Bacteroides,Prevotellaceae,Neisseriaceae and Helicobacteraceae represent,on average,more than 60% of the sequences found in each sample.2.In patients with chronic gastritis,H.pylori positive and H.pylori negative there was no significant difference in gastric microbiota.3.H.pylori has nothing to do with gender and age,but H.pylori positive rate increased with age.4.Non-atrophic gastritis patients had a H.pylori positive rate of 43.90%,CAG H.pylori positive rate was 72.88%,and CAG H.pylori positive rate was significantly higher than that of non-atrophic gastritis.5.In traditional Chinese medicine,The common TCM Syndromes of chronic gastritis are spleen-stomach sampness-heat,stagnation of blood-stasis in stomach,liver and stomach heat,spleen-stomach weak-coled,deficiency of stomach yin,liver and stomach qi stagnation,and spleen and stomach qi deficiency syndrome,of which spleen-stomach sampness-heat was the most common,accounting for 38.75%,spleen and stomach qi deficiency accounts for 21.25%,liver and stomach heat syndrome accounted for 15.00%,liver and stomach qi stagnation accounted for 12.50%,spleen-stomach weak-coled accounted for 7.50%,deficiency of stomach yin accounted for 3.75%,and stagnation of blood-stasis in stomach accounted for 1.25%.6.Among the TCM syndromes of chronic gastritis,the highest positive rate of H.pylori was the spleen-stomach sampness-heat,accounting for 90.32%;followed by liver and stomach heat,accounting for 83.33%,spleen-stomach weak-coled,liver and stomach qi stagnation,spleen and stomach qi deficiency account for 33.33%,30.00%,35.29%,respectively;stagnation of blood-stasis in stomach and deficiency of stomach yin were not infected with H.pylori.The H.pylori positive rate of spleen-stomach sampness-heat and liver and stomach heat were different from the syndromes of spleen and stomach qi deficiency,spleen-stomach weak-coled,spleen and stomach qi deficiency,stagnation of blood-stasis,deficiency of stomach yin;There was no difference between spleen-stomach weak-coled and spleen and stomach qi deficiency and spleen and stomach qi deficiency.Conclusion: The common gastric microbiota of patients with chronic gastritis were Streptococcaceae,Veillonellaceae,Bacteroides,Prevotellaceae,Neisseriaceae and Helicobacteraceae.There was no significant difference between H.pylori positive and H.pylori negative in gastric microbiota.H.pylori was closely related to CAG.In the traditional Chinese medicine syndrome of chronic gastritis,H.pylori was closely related to spleen-stomach sampness-heat and liver and stomach heat.These views provide an effective way to study the relationship between chronic gastritis and f gastric microbiota.
