Objective Bladder cancer is the most common malignant tumor in the urogenital system in China in terms of morbidity and mortality.It has a high recurrence rate,with a 5-year recurrence rate close to 50%.Therefore,bladder cancer has become a research hotspot at home and abroad.Yes-associated protein 1(YAP1),as a node gene of Hippo signaling pathway,play a vital role in the occurrence and development of epithelial tumors.The experiment is to study the expression of YAP1 protein in bladder cancer tissues and bladder cancer cells,and further study the effect of YAP1 protein on the proliferation,migration of bladder cancer cells.To study the antitumor effect of verteporfin(VP)in bladder cancer and to provide new strategies for clinical treatment of bladder cancer.Methods A total of 44 specimens of bladder urothelial cancer tissues,10 specimens of normal bladder mucosa tissue adjacent to the cancer were collected from the Second Hospital of Tianjin Medical University in this study.The expression level of YAP1 protein in bladder cancer tissues and adjacent normal bladder mucosa tissues was detected by immunohistochemical staining.The expression levels of YAP1 protein in human bladder cancer cells T24,5637,EJ and BIU-87 were detected by western-blot.We further tested the role of YAP1 protein in human bladder cancer 5637,EJ cell proliferation,migration through MTT,cloning experiments and Transwell.The cell scratch test and cloning experiment verified the inhibitory effect of Verteporfin on the proliferation and migration of human bladder cancer 5637 and EJ cells.Results Immunohistochemical staining results showed that the positive expression of YAP1 protein in bladder cancer tissues was prominently higher than that in adjacent normal bladder mucosa tissues(p<0.0001).Western-blot results showed that the expression of YAP1 protein was relatively high in human bladder cancer 5637,EJ,and BIU-87 cells.The results of MTT and cloning experiments show that YAP1 can significantly promote the growth and proliferation of human bladder cancer 5637 and EJ cells.Transwell experiment results show that YAP1 can significantly promote the migration of human bladder cancer 5637 and EJ cells.The results of cell scratching and cloning experiments show that vertepofen has a significant inhibitory effect on the proliferation and migration of human bladder cancer 5637 and EJ cells.Conclusion The expression of YAP1 protein in bladder urothelial carcinoma is significantly higher than that of normal bladder mucosa tissue adjacent to the cancer.YAP1 protein can promote the proliferation,migration of bladder cancer cells.Vertepofen can significantly inhibit the proliferation and migration of bladder cancer cells.
