The Preparation Of Hydroxysafflor Yellow A-loaded Hydrogel And Its Effect On Acute Wound Healing

Object:To investigate the therapeutic effect of hydroxysafflor yellow A(HSYA)on acute wounds.Prepare a topical delivery of HSYA based on gelatin-chitosan hydrogel as carrier,and explore its effect on promoting acute wound healing and related mechanisms.Methods: 1.The therapeutic effect of HSYA solution on skin wounds was investigated by a series of experiments in vitro.The effects of HSYA solution on the proliferation of the spontaneously immortalized human skin keratinocyte cell(Ha Ca T)and vascular endothelial cells(Endo)and the migration of human immortalized keratinocytes(Ha Ca T)were investigated.It was investigated that the effect of HSYA on angiogenesis via tube formation experiment.2.The determination of HSYA in hydrogels was developed by HPLC.The gelatin-chitosan hydrogel carrying HSYA was constructed.A series of studies were carried out on the weight loss rate,swelling ratio,rheological properties and cumulative drug release rate of hydrogels.Then determin the final formulation.3.In vivo experiments,the skin irritation and wound repair experiments of HSYA gelatin-chitosan hydrogel were investigated.The wound healing effect of HSYA gelatin-chitosan hydrogel was verified by Hematoxylin-eosin and Masson trichrome staining and VEGF immunohistochemical staining.Results:1.It was discovered that a strong effect proliferation on Endo and Ha Ca T by HSYA,which owns a certain effect on promoting keratinocyte migration and blood vessel formation.2.The gelatin-chitosan hydrogel was prepared by different concentrations of gelatin and chitosan according to a certain ratio after cross-linking by genipin.The prepared hydrogel shows suitable p H value,loss on drying weight,water absorption rate and suitable rheological properties,certain drug sustained release function,and is suitable for local application to the skin.It was determined that the final formulation content of gelatin-chitosan hydrogel for subsequent testing was 3% gelatin,8% chitosan,mass ratio 7:3.3.The prepared HSYA gelatin-chitosan hydrogel showed no skin irritation by the skin irritation test.In the wound healing experiment,HSYA gelatinchitosan hydrogel can improve the wound healing rate of skin wounds.The results of morphology and immunohistochemistry by examining the skin after healing showed that the HSYA gelatin-chitosan hydrogel can promote skin angiogenesis and the formation of skin accessory organs and the expression of VEGF,which showed a good healing effect.Conclusion: HSYA gelatin-chitosan hydrogel can be used as a good local drug delivery system for promoting wound healing.
