Study On The Chemical Constituents And The Quality Standards Of Sparganii Rhizoma

Sparganii Rhizoma is the dried tubers of Sparganium stoloniferum Buch.-Ham,It has the effects of breaking blood and qi,eliminating product and analgesic.It was reported that there were phenylpropanes,flavones,essential oils,organic acids,alkaloids and other chemical components in Sparganii Rhizoma.In this thesis,the chemical compositions and the quality evaluation of Sparganii Rhizoma were studied.It is a beneficial foundation to the follow-up study pharmacodynamic material basis,quality evaluation research and clinical application.Compounds were separated and purified by macroporous adsorption resin,silica gel column chromatography,gel column chromatography,preparative high performance liquid chromatography,and conventional recrystallization.Their structures were identified according to nuclear magnetic resonance,highresolution mass spectrometry,and physical and chemical properties,Twelve compounds were isolated and identified from the water layer and n-butanol layer of Sparganii Rhizoma.There were six cyclic dipeptides: cyclo(nbutyl-Glu-Phe)(1),cyclo(n-butyl-Asp-Phe)(2),cyclo(Glu-Phe)(3),cyclo(Leu Tyr)(4),cyclo(n-butyl-Glu-Leu)(11),cyclo(n-butyl-Glu-Val(12),two amino acids: phenylalanine(5)and pyroglutamate(6),1 alkaloid compound: 1,2,3,4-tetrahydro-1-methyl-carboline-3-carboxylic acid(9),and drynaran(7),3-hydroxyl-3-methyl glutarate(8),n-butyl oglucoside(10).cyclo(n-butyl-Glu-Leu),(1),(11)and(12)are new constituents.(2),(3),(5),(6),(7),(8)and(9)were isolated from this plant for the first time.The fingerprints of 14 batches Sparganii Rhizoma were established by HPLC,15 common peaks were obtained,similarity analysis showed that the similarity of 14 batches of Sparganii Rhizoma was between 0.663-0.937,and there was a big difference in composition content between the Sparganii Rhizoma in the market.And 8 chief peaks were determined as: 5-hydroxymethylfurfural,protocatechuic acid,vanillin,vanillic acid,caffeic acid,p-coumaric acid,ferulic acid,1-O-ferulic acid-3-p-coumaric acid.The content determination of 8 components by HPLC was established and the 14 batches Sparganii Rhizoma were determined.The contents of these samples were significantly different.The RSDs were sorted from high to low: 5-hydroxymethylfurfural,ferulic acid,caffeic acid,1-O-ferulic acid-3-p-coumaric acid,vanillin,p-coumaric acid,protocatechuic acid.To determine the content of the above 8 components of the three-size Sparganii Rhizoma,and their corresponding outer skin,fibrous root and petiole base from Hunan Huarong.The contents of protocatechuic acid,vanillin,p-coumaric acid,1-O-ferulic acid-3-p-coumaric glyceride in the petiole residue,tuber skin and fibrous root were higher than those in the medicinal materials.The research provides references for the harvesting and processing,improving the quality and further research of Sparganii Rhizoma.
