| Objective:To explore the comparison of the antihypertensive effect of Pueraria lobata Ohwi and Pueraria thomsonii Benth on L-NAME(N~ω-Nitro-L-arginine methyl ester)induced hypertensive rats and the possible mechanism.Methods:The experimental subjects were 90 healthy and clean male SD rats.Ten of them were selected as the normal group according to the random principle.Th e remaining 80 SD rats were modeled and intraperitoneal injected with L-name(25mg/kg)once a day for 8 weeks.The weight and blood pressure of the rats were measured once a week.After 4 weeks of modeling,the rats with NO val ue and systolic blood pressure≥140mm Hg were selected as hypertensive rats a nd random Ly divided into 8 groups:model group,captopril group,Pueraria lob ata Ohwi low concentration group,medium concentration group,high concentrat ion group,Pueraria thomsonii Benth low concentration group,medium concentr ation group,high concentration group.Plus the blank group,there are 9 group s.After successfμL modeling,hypertensive rats were given both intraperitoneal injection and gavage and kept for 4 weeks.The weight and blood pressure of the rats were measured weekly.After 4 weeks,blood was taken from the abdo minal aorta and heart,thoracic aortic and kidney tissues were extracted.Plasma concentrations of NO,e NOS,LDH,SOD and ANP were measured,and the h eart weight ratio was calcμLated by weighing.The histopathological changes of heart,thoracic aorta and kidney were observed by HE staining.Results:1.After 4 weeks of intraperitoneal injection of L-NAME,a hypertensive rat model was successfμLly made.The systolic blood pressure was significantly higher than before administration and compared with the normal group(P<0.001),and the detection of nitric oxide(NO)in the model group was significantly lower than that in the normal group(P<0.001);2.After gavage,the low,medium,and high concentration of Pueraria lobata Ohwi lowered blood pressure,and the systolic blood pressure began to decrease after 3weeks of administration compared with the model group(P<0.05),and decreased significantly after 4 weeks(P<0.01),and the systolic blood pressure of the Pueraria thomsonii Benth low,medium and high dose groups after 4 weeks of administration was not different from the model group.Captopril decreased the systolic blood pressure of the model group after 4 weeks and had statistical Significance(P<0.01);3.After taking the material,weigh the heart.Pueraria lobata Ohwi low,medium,and high concentrations reduced the heart weight ratio(P<0.01,P<0.001).Pueraria thomsonii Benth low,medium,and high dose groups did not significantly reduce the heart/weight value,and There was no statistical significance in the comparison of the model group,and captopril reduced the heart/weight value(P<0.05);4.Abdominal aorta blood was taken to detect plasma NO,LDH,and SOD concentrations.Pueraria lobata Ohwi and captopril groups increased NO and SOD levels and decreased plasma LDH levels,while Pueraria thomsonii Benth group did not.Enzyme-linked immunoassay(ELISA)Detecting the concentrations of ANP and e NOS,Pueraria lobata Ohwi and Captopril groups reduced ANP content and increased e NOS content,while Pueraria thomsonii Benth group did not change the above indicators;5.Histopathological changes showed that Pueraria lobata Ohwi improved rat myocardial hypertrophy,myocardial fibrosis and edema.Pueraria lobata Ohwi improved the swelling of vascμLar endothelial cells with rupture and proliferation of smooth muscle cells in the middle membrane,It improved the atrophy of glomerμLi and renal tubμLes,interstitial vascμLar congestion and mononuclear cell infiltration,etc.while radix puerariae had no significant changes in the heart,vascμLar and kidney tissue.Conclusion:1.Pueraria lobata Ohwi Pueraria root has therapeutic effect on L-NAME induced hypertension in rats,while Pueraria thomsonii Benth has no effect on hypertension.2.The mechanism of Pueraria lobata Ohwi for the treatment of hypertension is related to the improvement of e NOS and SOD activity and the regμLation of ANP secretion. |
