| ObjectiveIschemic stroke(IS)is a common and high incidence disease in clinic,which is closely related to the destruction of endothelial structure and function,In recent years,it has been found that endothelin-1(ET-1)plays an important role in the occurrence and development of ischemic stroke;Therefore,the relationship between ET-1 and is and the treatment of is with its antagonists are the research hotspot at present,but the research of traditional Chinese medicine on ischemic stroke and ET-1 is less.Ligustrazine is the main component of traditional Chinese medicine Ligusticum chuanxiong,which can promote blood circulation and remove blood stasis.It has the pharmacological effects of protecting endothelial cells,anti atherosclerosis and so on.It is widely used in the treatment of ischemic cerebrovascular disease.In this paper,we will use different concentrations of Ligustrazine to intervene is rats,and observe the relationship between endothelin-1 and ligustrazine,To provide experimental basis for the clinical application of ligustrazine in the prevention and treatment of ischemic stroke.MethodsThe clean SD rats,male,weighing(248.0 ±14.38)g,Totally randomly divided into 10 sham operation group(group C)and 30 model group,The models were divided into normal saline group(NS group),high Ligustrazine group(HT group),low Ligustrazine group(LT group),10 rats each.Before the experiment,all rats were fed adaptively in the central laboratory for one week,and were free to eat and drink water;One week later,the middle cerebral artery was embolized by thread embolism,In group C,only the carotid artery was separated,but not embolized;The rats were weighed every day after modeling,and the dosage of Ligustrazine and normal saline were calculated according to the weight,In LT group and HT group,ligustrazine injection was given intraperitoneally 50 mg/(kg.d)、75mg/(kg.d),Group C and NS were given equal volume saline for 4 weeks.The rats were weighed every day,and the dosage of Ligustrazine and normal saline were adjusted according to the weight;Rats in each group after modeling and treatment mNSS was used to evaluate the neural function of rats;Blood collection of orbital venous plexus in rats after 2 and 4 weeks of treatment,The content of ET-1 in serum was detected by ELISA.Result: 1 Comparison of neural function scores of MNSs in rats:Postoperative 24-hour score,The neurological score of the model group was higher than that of the sham operation group,(P<0.05),NS group was compared with HT group(P=0.63),compared with LT group(P= 0.44),compared with HT group(P=0.16),The difference was not statistically significant,It shows that the model of ischemic stroke in rats is successful and comparable.Neurological score 4 weeks after operation,C group(0.20±0.42),NS group(10.80±1.14),LT group(9.80±0.63),HT group(7.20±1.03),The neurological function score of HT group was lower than that of LT group(P<0.05),The high dose group was significantly lower than the low dose group,Tetramethylpyrazine can reduce the neurological score of rats,and the effect of high dose is more significant.2 Comparison of serum ET-1 levels in rats(1)Compared with group C,the content of ET-1 in the model rats increased(P<0.05),(2)Comparison of different dosage: The content of ET-1 in LT group was lower than that in NS group(P < 0.05),HT was significantly lower than NS group(P < 0.05),HT group was significantly lower than LT group(P < 0.05);Tetramethylpyrazine can reduce the content of serum ET-1 in is rats,and the high dose group is better than the low dose group.(3)Comparison of administration time: Compared with 2 weeks after administration,After 4 weeks,the serum ET-1 of HT group and lt group decreased(P<0.05),There was no significant difference between group C(P = 0.92)and group ns(P = 0.32).The effect of Ligustrazine on ET-1 in IS rats.ConclusionTetramethylpyrazine can significantly reduce ET-1 content and improve neurological deficit score in rats with ischemic stroke,It has a good therapeutic effect on ischemic stroke rats. |
