Objectives:Using the method of evidence-based medicine,systematically evaluate the clinical effect of traditional Chinese medicine on lung cancer.The data mining method is used to mine the prescriptions of Chinese medicine for lung cancer,analyze and summarize the rules of Chinese medicine for lung cancer,and provide clinical evidence and scientific research directions for the treatment of lung cancer by Chinese medicine.Using network pharmacological methods to predict the mechanism of the core drugs of Chinese medicine for lung cancer.Methods:(1)Meta analysis methodDevelop retrieval strategy,search literature in CNKI,CBM,VIP,Wanfang database,and incorporate into the randomized controlled trial and semi randomized controlled trial of TCM combined chemotherapy for lung cancer.Screen literature according to the inclusion and exclusion criteria,extract data,and use Rev Man5.3 software to perform Meta analysis.(2)Data mining analysis methodsThe clinical prescriptions of Chinese medicine for lung cancer were collected from the Affiliated Hospital of Jiangxi University of Traditional Chinese Medicine,and TCMISS was used to explore the combination of high-frequency drugs,performance distribution,strongly related drugs and core drugs commonly used in lung cancer,and to analyze the rules of Chinese medicine treatment of lung cancer.(3)Network Pharmacology MethodUsing TCMSP to retrieve the main components of the core drugs and their corresponding targets;using Malacards,CTD,TTD disease database to retrieve the related targets of lung cancer diseases.The intersection of the two results obtained by the Merge plug-in is used as a potential target for core drugs to treat lung cancer.Map the PPI network relationship through the String database and Cytoscape software,and perform network topology analysis to obtain key targets.Finally,go biological function and KEGG metabolic pathway of key nodes were analyzed by David database.Results:(1)In the Meta-analysis.a total of 40 controlled trials were included,including 439 patients,including 222 in the test group and 217 in the control group.Meta analysis results show that the total effective rate of the experimental group is significantly better than that of the control group(P<0.01);in terms of the incidence of adverse reactions,the experimental group is significantly better than the control group;compared with the changes in the immune function of the T cell population in the two groups,the experimental group is significantly better than Control group.(2)In the data mining of lung cancer prescriptions.a total of 164 prescriptions were included,and 310 Chinese medicines appeared,of which 125 were used more than 5 times;the performance distribution of the 125 traditional Chinese medicines before the statistics was calculated.Mainly "bitter,sweet,and astringent",attributed to "lung meridian,liver meridian,stomach meridian,spleen meridian";the top three drug combination models supported by association rule analysis are licorice-poria,atractylodes-poria,astragalus-Poria,Atractylodes — Licorice;the top three with confidence are Atractylodes,Licorice-> Poria,Chenpi-> Poria,Atractylodes-> Poria,Atractylodes,Poria-> Licorice.Entropy cluster analysis yielded 14 drug combinations and 7 new prescription combinations.(3)In the network pharmacology analysis.the 7 core drugs used oral bioavailability(OB value)and drug-like properties(DL value)as the screening criteria to obtain 101 active ingredients corresponding to 278 targets;the disease database screened 838 lung cancers target;By comparing the two results,97 potential targets for core drugs for lung cancer were obtained.Cytoscape was used to draw a visual map of protein interaction networks.Eight key targets such as EGFR,JUN,AKT1 were obtained through network topology analysis.GO function was enriched to 19 items,including 11 biological functions,3 cellular components,and 5 molecular functions;KEGG enrichment analysis screened the first 30 signal pathways.The enrichment results show that the core drugs may treat lung cancer by regulating the signal pathways such as PI3K-Akt,JAK/STAT,TGF-β,Pathways in cancer and Non-small cell lung cancer.Transduction,positive regulation of RNA polymerase II promoter transcription and other biological processes.Conclusion:(1)Chinese medicine combined with chemotherapy is more effective than chemotherapy alone in the treatment of lung cancer,and has obvious effects in reducing adverse reactions and has higher clinical application value.(2)Data mining results show that the traditional Chinese medicine treatment of lung cancer chemotherapy patients mostly adopts the method of cultivating soil and generating gold.The treatment is mainly to invigorate Qi and Yin,supplemented by clearing heat and detoxification,promoting blood circulation and removing blood stasis,soft and hard powder.(3)The therapeutic effect of core drugs on lung cancer embodies the characteristics of multi-component,multi-target and multi-channel of traditional Chinese medicine,which provides experimental research basis and ideas for further exploring the mechanism of traditional Chinese medicine in treating lung cancer. |
