| Objective:the effects of Xiezhuo Qingjie Decoction on the general situation,disease activity index(DAI),pathological changes of colon tissue,expression of myeloid differentiation factor 88(My D88)in colon tissue and content of interleukin-18(IL-18)in serum of Wistar rats with ulcerative colitis model were observed,and the mechanism of Xiezhuo Qingjie Decoction in treating ulcerative colitis(UC)was discussed,providing experimental and theoretical basis for clinical application of Huazhuo Qingjie Decoction in treating UC.Method:Sixty healthy Wistar rats were randomly divided into 6 groups with 10 rats in each group,namely blank control group(NG),model group(MG),western medicine mesalazine group(MS),Xiezhuo Qingjie decoction high dose group(CMH),medium dose group(CMM)and low dose group(CML).Except for normal feeding and no treatment in NG group,all other groups were modeled by TNBS/alcohol compounding method.After 2 days of modeling,2 rats were randomly selected and killed.Colonic tissues were dissected and obtained.After being washed with physiological saline,macroscopic observation showed hyperemia and edema of colonic mucosa with ulcer formation.Ask a pathologist to assist in diagnosis and confirm the success of modeling.On the 4th day after successful modeling,the MG group and each treatment group were given normal saline and corresponding drugs for gastric perfusion.After continuous intervention for 14 days,the general conditions of rats were observed and DAI score was made.After the treatment,the rats were killed,blood was taken from femoral artery,colon tissue with obvious pathological changes was dissected and taken,expression of My D88 in intestinal mucosa tissue and serum IL-18 content were detected.Results:1.General situation and DAI scoreDuring the experiment,the NG group rats were in the best general condition,with rapid movement,sensitive response,bright and soft fur,feeding preference,normal diet,weight gain,black and granular stool,and clean crissum.The other groups of rats showed listlessness after modeling,preferring to huddle up and lie down,messy and lusterless fur,not preferring to feed,emaciation,increased stool frequency in different degrees,thin quality,accompanied by mucus,pus,bloody stool and perianal filth.After drug intervention,the general situation of rats in MS group,CMH group,CMM group and CML group has been improved to different degrees,of which CMH group has the most obvious effect,close to NG group.There was no significant difference in improvement between CMM group and MS group,which was worse than CMH group.The CML group had the worst improvement,while the MG group had worse general conditions,such as listlessness,loss of appetite,perianal filth,increased mucus,pus and blood,and weight loss.DAI score resultsThe scores of CMH,CMM,CML and MS groups are lower than those of MG group.The score of CMH group is lower than that of CMM,CML and MS groups.The scores of CMM and MS group are lower than those of CML group.The above differences were statistically significant(P<0.05).There was no significant difference between CMM group and MS group(P>0.05).2.Macroscopic and pathological observations of colon tissueColonic tissue of NG group rats was normal,no edema or ulcer was found.Colonic epithelium was intact and crypts were arranged orderly under microscope,with a large number of goblet cells visible.Colonic erosion and ulcer formation were observed in MG group rats.The crypt branches,distortion and atrophy were observed under microscope,accompanied by a large number of inflammatory cell infiltration.The macroscopic and microscopic observation of colon in CMH rats is close to that in NG group.CMM group is similar to MS group,intestinal mucosa is less smooth,inflammatory cell infiltration can be seen under microscope,goblet cells are more,which is better than CML group and worse than CMH group.In CML group,the intestinal mucosa was rough,mucosal hyperemia could be seen sporadically,epithelial defects were obvious under microscope,crypts were irregularly arranged,and more inflammatory cells infiltrated.3.Index detectionImmunohistochemical results of colonic tissue My D88:The expression level of My D88 in NG group was lower than that in other groups;The expression level of My D88 in CMH group was lower than that in MG,MS,CMMM and CML groups;The expression level of My D88 in CMM and MS groups was lower than that in MG and CML groups;The expression level of My D88 in CML group was lower than that in MG group,the above differences were statistically significant(P<0.05).There was no significant difference in the expression level of My D88 between CMM and MS group(P>0.05).Serum IL-18 ELISA results:The content of serum IL-18 in NG group was lower than that in other groups.The content of serum IL-18 in CMH group was lower than that in MG,MS,CMM and CML groups.The content of serum IL-18 in CMM and MS groups is lower than that in MG and CML groups.The content of serum IL-18 in CML group was lower than that in MG group,and the above differences were statistically significant(P<0.05).There was no significant difference in serum IL-18 content between CMM and MS group(P>0.05).Conclusion:Xiezhuo Qingjie Decoction can improve the general condition of ulcerative colitis rats,reduce DAI score,and reduce colon mucosa damage.The mechanism of action may be to reduce the expression of My D88 in colon tissue and reduce the content of serum IL-18,reduce inflammatory reaction and improve immune function. |
