Study On Substance Benchmarks And Modern Technology Of Taohe Chengqi Decoction

Objective:To establish the substance benchmarks of the classical prescription Taohe Chengqi Decoction,and to select the best extraction process and the molding process of the granules to lay the foundation for the development of the classical prescription formulation.Methods: 1.Establishment of substance standard of Taohe Chengqi Decoction: 10 batches of substance benchmarks were prepared according to the original records of Taohe Chengqi Decoction.The paste ratio was measured,a fingerprint was established,a common peak was identified,and similarity evaluation was performed.The contents of amygdalin,rhein,emodin,chrysophanol,aloe-emodin,cinnamaldehyde,glycyrrhizin and glycyrrhizic acid were determined.2.Selection of optimum extraction process of Taohe Chengqi Decoction:Based on single factor investigation,combined with response surface methodology and information entropy theory,taking the contents of amygdalin,rhein,cinnamaldehyde,and glycyrrhizic acid as evaluation indicators,Study the soaking time,material-liquid ratio,and total extraction time to determine the optimal extraction process;Selection of optimum molding process of Taohe Chengqi decoction granules: On the basis of a single factor,an orthogonal test was performed to remove the paste density,paste and auxiliary materials the ratio,the ratio of powdered sugar to dextrin are the influencing factors.The content of the effective ingredients in the granules,the qualification rate and the solubility are used as indicators to determine the optimal molding process.Results: 1.The substance benchmarks paste rate of the 10 batches of Taohe Chengqi Decoction was 15.87 to 17.82%.The fingerprints identified 18 common peaks,and the similarity between the 10 batches was above 0.98,indicating that the differences between the batches were small and stable.2.The best extraction process of Taohe Chengqi Decoction was as follows:soaking time 60 min,material-liquid ratio 1:10(g / m L),total extraction time130 min(3 extractions,65,33,32 min each time));The best molding process of Taohe Chengqi Decoction granuleswas as follows: extract density 1.4,extract: auxiliary materials = 1: 4,powdered sugar: dextrin = 1: 3.Conclusion: The fingerprint method comprehensively reflects the composition characteristics of Taohe Chengqi Decoction,which can be used as the basis for material benchmark evaluation;The optimum extraction process and molding process selected are stable and reliable,which can provide ideas and research for the classic famous prescription.
