Application And Comparative Study Of IHC And QDB In Detection Of HER2 Protein In Breast Cancer

Objective:To compare the consistency of HER2 protein expression in breast cancer detected by immunohistochemistry(IHC)and Quantitative Dot Blot(QDB)and explore two detection methods.Methods:Data from 239 breast cancer patients admitted to Yantai Affiliated Hospital of Binzhou Medical College from January 1,2008 to December 31,2013 were collected,and the expression levels of HER2 protein in breast cancer tissues were detected and analyzed by IHC and QDB methods Its correlation with clinicopathological characteristics(age,tumor size,tumor location,lymph node metastasis,histological grade,TNM stage);the consistency of the two detection methods was evaluated by Kappa test.Five cases were randomly selected from the two test methods with inconsistent results.Fluorescence in situ hybridization(FISH)method was used for testing to further explore the accuracy of IHC and QDB methods.Results:IHC and QDB showed no significant correlation between HER2 protein expression and age,TNM stage,grade,tumor size,tumor location and lymph node metastasis in breast cancer tissues(P>0.05).Among the 239 breast cancer patients,169 cases(70.71%)had low HER2 expression and 70 cases(29.29%)had high expression by immunovision Eli vision method,and 184 cases(77.00%)had low expression of HER2 by QDB method.Cases(23.00%).Among them,162 cases with low expression and 48 cases with high expression were detected using these two methods.The consistency test showed that the Kappa value was 0.687,and the consistency between the two was relatively high.The FISH results of 5 specimens inconsistent with the results of the two detection methods were consistent with the QDB results.Conclusion:QDB and IHC have a high degree of consistency,and may have higher accuracy than IHC method,and can play an important role in the detection of clinical breast cancer HER2 protein.
