| Objective:The influence of host genetic factors on vaccine induced antibody response is an important direction of vaccine genomics.This study aims to explore the possible effect of MHC related factors on the response of polio antibody induced by sequential immunization of polio vaccine by analyzing the single nucleotide polymorphism(SNPs)of MHC I antigen presenting pathway gene,including low molecular weight proteasome gene(LMP),transporter associated with antigen processing gene(TAP)and endoplasmic reticulum aminopeptidase gene(ERAP).Methods:357 individuals,2-3 years old,were selected from Guangxi Zhuang Autonomous Region who received 2-dose of inactivated polio vaccine(IPV)and 1-dose of bivalent oral polio vaccine(bOPV).The serum levels of neutralizing antibodies to three types of poliovirus in each participants before vaccination and 28 days after basic immunization were tested.Using the TaqMan probe genotyping method,LMP2 gene rs17587(G>A),LMP7 gene rs2071543(G>T),TAP1 gene rs41549617(G>A),rs1057141(C>T),rs1135216(T>C),rs1057149(C>T),TAP2 genes rs1042116(G>A),s2228396(C>T),rs4148876(G>A),ERAP1 genes rs27037(G>T),rs27044(C>G),rs30187(C>T),rs26618(T>C),rs26653(C>G),rs3734016(C>T),ERAP2 gene rs2549782(T>G),rs2548538(A>T),rs2248374(G>A),rs2287988(A>G),rs1056893(T>C)sites were genotyped,the allele and genotype frequencies of each SNPs site were calculated,and the correlation between SNPs and polio antibody response was analyzed.Results:(1)There was no significant difference in the GMT levels of neutralizing antibodies among the three types of polio vaccine induced by LMP2 gene rs175 87,LMP7 gene rs2071543,TAPI gene four SNPs,TAP2 gene three SNPs;There was no difference in the level of GMT among the carriers with different TAP1 alleles and TAP2 alleles.(2)The GMT of neutralizing antibody of rs3734016-C allele of ERAP1 gene was lower than that of T allele(11.914±0.074 vs 12.375 ± 0.166,P=0.024:6.093±0.072 vs 6.548 ±0.188,P=0.025).There are differences in GMT levels of neutralizing antibodies against poliovirus type I among carriers of each genotype at the rs2549782、rs2548538、rs2248374、rs2287988、rs1056893 locus of the ERAP1 gene(11.9251±0.223 vs 11.741 ± 0.141 vs 12.378 ± 0.157,P=0.016,P_adj>0.05),but there was no statistical difference after Bonferroni correction.(3)LMP,TAP,ERAP SNPs did not show the correlation between single or combined action and polio type Ⅱ antibody positive conversion(P>0.05).Conclusion:LMP,TAP,ERAP gene polymorphisms did not show significant correlation with antibody response after immunization. |
