MMP1/MMP3 Regulated By UBE3A Affects Proliferation,Invasion And Migration Of Triple Negative Breast Cancer | | Posted on:2022-10-22 | Degree:Master | Type:Thesis | | Country:China | Candidate:J J Li | Full Text:PDF | | GTID:2504306347487234 | Subject:Human Anatomy and Embryology | | Abstract/Summary: | PDF Full Text Request | | Objective:This study explore the correlation between the expression of MMPland MMP3 in breast cancer and their effects on proliferation,invasion and metastasis of breast cancer cells with their mechanism and explore the relationship between UBE3A and MMP1/MMP3 in breast cancer.Methods:The expression of MMP1 and MMP3 in breast cancer and adjacent normal tissues was detected by immunohistochemistry,their correlation was analyzed.MMP1 was knockeddown and overexpressed in breast cancer cell MDA-MB-231.To explore the change of biological behavior,plate cloning assay was used to observe the change of cell proliferation,Transwell chamber assay was used to observe the invasion and migration ability of breast cancer cell MDA-MB-231.The stable transfection cell model was constructed with lentiviralvector knockdown/overexpression MMP1 in MDA-MB-231.The change of UBE3A was observed after MMP1/MMP3 knockdown in breast cancer cell MDA-MB-231,the changes of MMP1/MMP3 were observed after UBE3A knockdown,construct the stable transfection cell model,the efficency of MMP1 knockdown and overexpress were observed by qPCR and Western blot and the efficency of knockdown MMP3 and UBE3A were detected.Results:The expression of MMP1 and MMP3 in breast cancer tissues was significantly higher than that in adjacent tissues(both P<0.01).After knockdown MMP1,the expression level of MMP3 increased,the ability of cell cloning decreased,the ability of cell invasion and migration enhanced,the level of MMP3 expression decreased after overexpression of MMP1,the ability of cell cloning decreased,the ability of invasion and migration decreased.PCR detected that the expression of MMP1 and MMP3 decreased after knockdown UBE3A,Western blot detected that the expression of MMP3 decreased after MMP1 was knockeddown,the expression of MMP3 decreased after UBE3A was knockeddown.PCR assay showed that the expression of UBE3A decreased after MMP1/MMP3 knocked down.It is suggested that UBE3A may be the up stream factor of MMP1 and MMP3 which can regulate the expression of MMP1/MMP3.Conclusions:MMP1,MMP3 and UBE3A are highly expressed in breast cancer,MMP1 may be the upstream regulatory factor of MMP3,MMP1 may negatively regulate the expression of MMP3,UBE3A is closely related to MMP1 and MMP3,they play an important role in the occurrence and development of breast cancer. | | Keywords/Search Tags: | Breast Cancer, MMP1, MMP3, UBE3A, Proliferation, Invasion and Migration | PDF Full Text Request | Related items |
