Objective:To compare the expression of Embryonic Lethal Abnormal Vision Drosophila Kike 4(ELAVL4,HuD),growth associated protein-43(gap43),and p21 among seborrheic keratosis(SK)lesions,skin around SK lesions,normal skin of different ages,to compare it in the blood of normal people,to investigate the role of HuD,gap43 and p21 in SK and skin aging;compare the expression of HuD related gene(p53 mRNA)and non-coding gene(lincJPX)in the blood of normal people of different ages,to explore whether they are related to aging.Methods:The specimens of patients who were treated at Affiliated Hospital of Yangzhou University from September 2019 to August 2020 were selected.Mainly include 25 wax block specimens,85 fresh skin samples and 54 blood samples.Compare the expression of HuD in SK lesions,skin around SK lesions,normal skin of different ages and the blood of normal people of different ages by Immunohistochemistry,Western blot and real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-PCR);compare the expression of the downstream proteins of HuD(gap43,p21)in SK lesions and normal skin by Western blot;compare the expression of HuD related gene(p53 mRNA)and non-coding gene(lincJPX)in the blood of normal people of different ages by RT-PCR.Results:1.IHC results:1)Compare the expression of HuD among normal skin of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.HuD is mainly expressed in basal layer,and the expression of HuD showed an increasing trend with age(P<0.01).2)Compare the expression of HuD among SK lesions,skin around SK lesions andnormal skin at the same age:The expression of HuD in SK lesions was higher than that in skin around SK lesions and normal skin(P<0.001).2.Western Blot results:2.1 Comparison of the expression of HuD1)Compare the expression of HuD among normal skin of different ages:It is divided into three groups:20-39 years old,40-59 years old,and≥60 years old,the expression of HuD show an increasing trend with age(P<0.05).2)Compare the expression of HuD between normal skin and SK lesion at the same age:The expression of HuD in seborrheic keratosis was higher than that in normal skin(P<0.05).3)Compare the expression of HuD between SK lesions and the skin around SK lesions at the same age:The expression of HuD in SK lesions was higher than that in skin around SK lesions(P<0.01).2.2 Comparison of the expression of GAP43 and P211)Compare the expression of GAP43 between normal skin and SK lesion at the same age:The expression of GAP43 in SK lesions was lower than that in normal skin(P<0.05).2)Compare the expression of p21 between normal skin and SK lesion at the same age:The expression of p21 in SK lesions was lower than that in normal skin(P<0.05).3)Compare the expression of p21 among normal skin of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.The expression of p21 in the three groups has no obvious trend with age(P>0.05).3.RT-PCR results:1)Compare the expression of HuD mRNA among the normal skin of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.The expression of HuD mRNA show an increasing trend with age(P<0.05).2)Compare the expression of HuD mRNA among the blood of normal people of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.The expression of HuD mRNA show an increasing trend with age(P<0.01).3)Compare the expression of P53 mRNA among the blood of normal people of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.The expression of p53 mRNA decreased in≥60 years old group,there was a statistically significant between the group of 40-59 years old and the group of≥60 years old(P<0.05).4)Compare the expression of lincJPX among the blood of normal people of different ages:It is divided into three groups:20-39 years old,40-59 years old and≥60 years old.The expression of lincJPX in the three groups has no obvious trend with age(P>0.05).Conclusion:The expression of HuD in SK lesion was higher than that in the skin around SK lesions and normal skin,the expression of HuD in normal skin and the blood of normal people increased with age,HuD may be associated with seborrheic keratosis,and HuD may be age-related..The expression of GAP43 and P21 in seborrheic keratosis was lower than that in normal skin,HuD may play a role in seborrheic keratosis through the signal pathway mediated by GAP43 or P21.The expression of p53 mRNA in the blood of normal people of 60 years old was decreased,it is verified that p53 is related to aging.
