Objective: To investigate the expression of Th1,Th2,Th17 cytokines,mi RNA-155 and mi RNA-21 in patients with different stages of brucellosis,and to understand the immune responses of brucellosis.Methods: Blood samples were collected from 125 patients with acute and chronic brucellosis and 56 healthy controls in different medical institutions in Xinjiang from July 2019 to November 2020.CBA was used to detect the expression levels of Th1,Th2 and Th17 cytokines in peripheral blood of patients with different stages of brucellosis and healthy controls.RT-PCR was used to detect the expression of mi RNA-155 and mi RNA-21 in plasma samples of patients with different stages of brucellosis and healthy controls.ESR and CRP test results were collected.Finally,the above results are analyzed.Results: The detection results of IL-18 and IL-33 in acute,chronic and control groups showed that the expression levels of IL-18 and IL-33 in acute and chronic groups were increased.The level of IL-18 in acute group was increased,and the level of IL-33 in chronic group was increased.IL-18 and IL-33 levels decreased after treatment.Further combined detection of Th1,Th2,and Th17 cytokines showed that the expression levels of Th1 cytokines(IFN-γ,TNF-α,and IL-18)in the acute and chronic groups were increased,and those in the acute group were significantly increased.The expression levels were decreased after treatment.The expression of Th2 cytokines(IL-10,IL-33)increased in both acute and chronic groups,and those in the chronic group were significantly increased.The expression of Th17 cytokines(IL-17)in the acute and chronic groups was increased,which was significantly increased in the acute phase,and decreased after treatment.The mi RNAs detection results showed that the expressions of mi RNA-155 and mi RNA-21 in the acute and chronic groups were increased,and the expression of mi RNA-155 was significantly increased in the acute group(all P < 0.05).Correlation analysis showed that the expression of IL-18 was positively correlated with the expression of IFN-γ and TNF-α.IL-33 expression was positively correlated with IL-10 expression;The expression of mi RNA-155 was positively correlated with the expression of Th1 and Th17 cytokines,and was positively correlated with CRP and ESR.Conclusion:Th1cytokines(IFN-γ,TNF-α,IL-18),Th2 cytokines(IL-10,IL-33)and Th17 cytokines(IL-17)are involved in the occurrence and development of brucellosis.In the acute phase,Th1 and Th17 cytokines were mainly expressed while in the chronic phase,Th2 cytokines were mainly expressed.The expression trend of IL-18 and IL-33 can reflect Th1 and Th2 immune effects respectively.mi RNA-21 and mi RNA-155 are involved in the body immunity after brucellosis infection,and mi RNA-155 is mainly expressed in the acute phase.mi RNA155 was significantly correlated with Th1,Th17 cytokines and brucellosis inflammation levels(CRP,ESR).In brucellosis,mi RNA155 may have a certain regulatory relationship with Th1 and Th17 cytokines.
