| Objective:By detecting the expression level of serum chemerin in intermittent hypoxia model of mice,the correlation between chemerin and metabolic syndrome factor was analyzed to explore the role of chemerin in the pathophysiological mechanism of OSAHS.Method:Twenty-eight healthy C57 male mice were randomly divided into four groups:normoxic+normal diet group(NC+ND),normoxic+high-fat diet group(NC+HFD),intermittent hypoxia+normal diet group(IH+ND),intermittent hypoxia+high-fat diet group(IH+HFD),n=7,respectively.The normal diet group was fed with basic diet and the high-fat diet group was fed with high-fat diet.The intermittent hypoxia group was placed in the intermittent hypoxia chamber for 8 hours a day,and the normoxic group was placed in the normoxic environment for 1 month.The body weight,liver weight,white fat weight and brown fat weight of mice were sampled and weighed;serum triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),free fatty acid(FFA)and fasting blood glucose were measured by automatic biochemical analyzer;serum chemerin expression and fasting insulin level were measured by ELISA method.Insulin resistance index(HOMA-IR)was used to calculate the degree of insulin resistance.Results:1.The body weight and liver weight of mice in the high fat group were significantly higher than those in the normal diet control group(P<0.05).The body weight and liver weight in the intermittent hypoxia group were significantly lower than those in the normoxic control group(P<0.05);2.The expression level of serum chemerin in IH+ND group was higher than that in NC+ND group,and the expression level of chemerin in NC+HFD group was significantly higher than that in NC+ND group(P<0.05);3.The serum levels of HDL-C,TC,TG and FFA in IH+ND group were higher than those in NC+ND group,the levels of HDL-C,TC,TG,FFA and LDL-C in NC+HFD group were higher than those in NC+ND group,the levels of TG and FFA in IH+HFD group were higher than those in NC+HFD group,and the serum levels of HDL-C,TC,TG,FFA and LDL-C in IH+HFD group were higher than those in IH+ND group;4.Pearson analysis showed that the level of serum chemerin in intermittent hy poxia group was positively correlated with body weight(r=0.8363,p=0.0190),white adipose tissue mass(r=0.8962,p=0.0063),brown adipose tissue mass(r=0.8610,p=0.0128)and TG(r=0.9082,p=0.0047),and negatively correlated with fasting insulin(r=-0.8488,p=0.0157)and insulin resistance(r=-0.8549,p=0.0142).Conclusions:The expression of serum chemerin in intermittent hypoxia mice tends to increase,and there is a disorder of blood glucose and blood lipid metabolism,which is more serious when combined with high-fat diet,and the change of serum chemerin level in intermittent hypoxia mice is related to the main components of metabolic syndrome,which plays a role in the development of OSAHS. |
