Object:To study the chemical composition,quality analysis,anti-Inflammatory and antioxidation effect of Zhuang Medicines Embelia Parviflora Wall n order to better develop and use of medicinal resources to do the preparation,to provide the scientific basis for follow-up of quality standards,preparation,clinical research.Methods:(1)Chemical compositions of research:Use silica gel colu-mn chromatography and LC-MS to analyze the chemical compositi on of Emibelia Parviflora Wall.,also analyze the different habitats of thevolatile oils by GC-MS.(2)Quality analysis:EmbeliaParviflora Wall.were identified by TLC,determine the total polysaccharides content by UV-Vis,and design for optimizing the optimum extraction for Box-Behnken central composite experiment,and then determination of its four kinds of active ingredients by HPLC,infrared spectral and cluster analysis were used to of Embelia parviflora Wall.from different habitats and different parts,GC-MS fingerprints analyze of Embelia parviflora Wall.From different habitats of the volatile oils.(3)Efficacy screening of research:the experimental study of anti-Inflammatory and antioxidation on Embelia parviflora Wall..Results:(1)Gallic acid,Protocatechuic acid,Catechin and Epicatechin were identified by LC-MS.124 components were isolated and identified from essential oil of different habitats of Embelia parviflo-ra Wall.,accounting for less than 60%of the total essential oil;(2)D-Glucose anhydrous spots clear and the corresponding position on the reference drug chromatogram was the same color spots in TLC;solid-liquid ratio was 32:1 ml/g,extraction temperature and extraction time were of 77℃ and 36 min.Good linear correlations were found in the ranges of 0.0896~0.7168、0.0966~0.8694、3.3134~23.1938、1.4424~12.9816μg/ml,for Gallic acid,Protocatechuic acid,Catechin and Epicatechin,respectively.The correlation coefficients were good.The averrage recoveries,respectively,the accuracy precision and stability were satisfactory with RSD of less than 2%.FTIR spectra of Codonops is Radix from different habitats had 9 common peaks.it were divided into four categories by using cluster analysis.FTIR spectra of Codonops is Radix from different parts had 6 common peaks.it were divided into two categories by using cluster analysis.There were 19 common peaks in GC-MS fingerprint of Embelia parviflora Wall.from different habitats of the volatile oils,the similarity of fingerprint were all higher than 0.9.(3)Embeli-ra Parviflora Wall.from different parts can inhibit the generation of inflammatory fator,like NO、TNF-α and IL-6 in a dose dependent manner.The n-butanol fractions IC50 values of DPPH and ABTS are lower than 0.04 mg/ml.The IC50 values of DPPH and ABTS are 0.04 and 0.03mg/ml of Polysaccharides from Embelia parviflora Wall..Conclusion:(1)The chemical composition analysis of Embelia parv-iflora Wall.was carried out to providea basis for the in-depth stud-y of Embelia parviflora Wall..(2)UV-Vis,infrared spectrum,HPLC performance chromatography and gas chromatography were used to control the quality of Embelia Parviflora Wall.in this experiment.The method is simple,accurate,stable and repeatable.(3)Anti-inflamma-tory effect of Embelia Parviflora Wall.isrelated to inhibit the production of inflammatory factors,like NO,TNF-α and IL-6,the n-butanol fractions have strong antioxidantactivity. |
