| BackgroundPeriostin is supposed to be a fetal protein,which effects the morphogenesis and development of bone and periodontal tissues.Distraction osteogenesis(DO)is used to treat limb-length discrepancies,bone deformities,and bone loss secondary to trauma,infection,or tumors.However,there are few reports about periostin during DO.Therefore,this study used three different distraction rates and intended to establish a rabbit mandibular DO model which produced poor bone regeneration.Four different concentrations of periostin were injected into the distraction gaps of the rabbit mandibular DO model.To study the effect of periostin during DO,all animals from each group were taken to imaging examination,histology and three-point bend testing after DO.Materials and Methods1.To established a rabbit mandibular DO model which produced poor bone regenerationWe used three different rates,including 0.5mm/d,1.0mm/d and 2.0mm/d,to accomplish unilateral DO in rabbits’ mandibular.Four weeks after DO,all animals from each groups were taken to scan by CT,X-ray examination,histology and threepoint bend testing.2.The study of periostin promoting rapid distraction osteogenesis in the rabbit mandibular model0.5ml of normal saline contained 0μg/kg,5μg/kg,10μg/kg,20μg/kg periostin was severally injected into the distraction gaps of DO model which produced poor bone regeneration.All animals from each groups were taken to scan by CT,DEXA examinations four weeks after DO and micro-CT,histology and three-point bend testing eight weeks after DO.Results:1.To established a rabbit mandibular DO model which produced poor bone regenerationIn this study,we used different distraction rates to accomplish rabbit mandibular DO.And we successfully established a rabbit mandibular DO model which produced poor bone regeneration.2.The study of periostin promoting rapid distraction osteogenesis in the rabbit mandibular modelIn this study,we injected different concentration of periostin into the gaps of the rabbit mandibular DO model which produced poor bone regeneration.Four weeks after DO,there were significant differences of imaging examinations between four groups of animals.Eight weeks after DO,the bone mineral density(BMD),bone mineral content(BMC)and the results of micro-CT of group C were apparently higher than others.And the three-point bending test demonstrated that the peak load of the distraction gaps of group C was the largest among four groups.(P<0.05).Conclusion:1.Contrasted with the distraction gaps of the normal distraction rate,more fibrous tissues were observed in the distraction gaps of the rapid distraction rate,which proved that poor bone regeneration could be produced under the rapid distraction rate in DO.2.Contrasted with the control group,groups with periostin achieved better bone regeneration.And the group contained 10μg/kg periostin was accelerated most strongly.It showed that although DO could be promoted by local application of periostin,the promotion was not in direct proportion to the concentration of periostin. |
