| Objective:(1)To explore whether Qufeng Xuanbi recipe can improve airway remodeling of asthma by regulating MEK/ERK signal pathway,so as to achieve the purpose of prevention and treatment of asthma.(2)Through the study of this subject,to provide molecular biological basis for Qufeng Xuanbi recipe in the treatment of airway remodeling of asthma.Methods:(1)In vivo experiment,18 SD male rats were randomly divided into 3 groups:control group,asthma group and Qufeng Xuanbi recipe group.The rat model of asthma was established by OVA,and the Qufeng Xuanbi recipe group was treated with Qufeng Xuanbi recipe for 2 weeks.The pathological changes of lung tissue were observed by HE staining,and IgE,IL-6 and IL-13 in serum and bronchoalveolar lavage fluid of rats were detected by ELISA to verify the therapeutic effect of asthma model and Qufeng Xuanbi recipe on asthmatic rats.(2)The level of airway remodeling was evaluated by detecting the levels of PCNA andα-SMA in the lung tissue of rats in each group,and the changes of MEK/ERK signaling pathway related molecules were detected to explore the mechanism of asthma airway remodeling and the regulatory intervention effect of Qufeng Xuanbi recipe.(3)The mouse airway smooth muscle cell AMSCs was used as the research object in vitro.The abnormal proliferation model of mouse airway smooth muscle cells induced by PDGF was used.Qufeng Xuanbi recipe or U0126,an inhibitor of MEK/ERK signal pathway,was used to interfere with the cells.Detect the changes of airway remodeling related indexes PCNA,α-SMA and related molecules of MEK/ERK signal pathway,and further verify that Qufeng Xuanbi recipe can improve airway remodeling of asthma by intervening MEK/ERK signal pathway,so as to achieve the purpose of prevention and treatment of asthma.Results:(1)OVA can induce inflammatory cell infiltration in lung tissue,increase serum IgE,IL-6 and IL-13,and increase IL-6 and IL-13 in bronchoalveolar lavage fluid.Qufeng Xuanbi recipe can inhibit pathological changes and improve airway inflammation in asthmatic rats.(2)Compared with the control group,the levels of PCNA and α-SMA in lung tissue of asthmatic rats increased,and Qufeng Xuanbi recipe could reduce the levels of PCNA and a-SMA in lung tissue of asthmatic rats.The protein phosphorylation level of MEK1/2 and ERK1/2 in lung tissue of asthmatic rats was higher than that of control group.Qufeng Xuanbi recipe could inhibit the phosphorylation level of MEK1/2 and ERK1/2 protein in lung tissue.(3)PDGF could significantly increase the expression of PCNA and α-SMA and the phosphorylation level of MEK1/2 and ERK1/2 in AMSCs.PDGF could inhibit the protein phosphorylation levels of PCNA,α-SMA,MEK1/2 and ERK1/2 after intervention with Qufeng Xuanbi recipe and U0126.Conclution:Qufeng Xuanbi recipe has a clear therapeutic effect on asthmatic rats.Regulating MEK/ERK signal pathway to improve airway remodeling is one of the mechanisms of Qufeng Xuanbi recipe in the treatment of asthma. |
