Objective: 1.To establish a rat model of functional constipation in the-10℃cryogenic freezer environment by simulating the transport and operation rules of the army from warm area to cold area,and to explore the pathogenesis of functional constipation in rats caused by low temperature environment by observing the detection indexes;2.To compare the prevention and treatment effects of Cistanche high dose group,medium dose group,low dose group and mosaprigo citrate tablet group on functional constipation in low temperature environment,and to develop the best drug regimen;3.By retrospectively analyzing 1 case of functional constipation caused by low temperature environment and collecting relevant data,more methods of clinical prevention and treatment of functional constipation were proposed.Methods: 1.Established in frozen ark-10 ℃ low temperature environment under the induction of rats with functional constipation model,through calculating the rat intestinal propulsion rate,detection SP content in serum of rats,indexes of AQP3 protein expression in the tissues of the colon,observing rats colon tissue pathological changes,so the low temperature environment lead to the pathogenesis of functional constipation in rats;2.According to the comparison of each index of rats in the high dose group,medium dose group,low dose group and mosaprigo citrate group,the prevention and treatment effect of Cistanche deserticola on functional constipation in low temperature environment was further studied.Results: 1.General situation: during intravagation,the defecation quantity of rats in Cistanche deserticola high-dose group increased,and the stool was softer than that in other groups,but there was no significant difference in other groups;Compared with blank group,6h after cold exposure,the defecation volume of rats in each group was significantly reduced,among which the model group had the least defecation volume,and the middle and high dose groups of C.deserticola had the most defecation volume,among which the high dose group of C.deserticola had the most defecation volume.2.Intestinal propulsion rate: Compared with the blank group,the intestinal propulsion rate of the model group was decreased;Compared with model group,intestinal propulsive rate was increased in low,medium and high dose Cistanche deserticola groups and mosapride groups.3.Intestinal neurotransmitter(SP):Compared with blank group,SP content in model group was decreased;Compared with the model group,SP content in the high dose group of Cistanche deserticola increased,but there was no significant difference between the low dose,medium dose and mosapride groups and the model group.4.Aquaporin(AQP3): Compared with blank group,the expression level of AQP3 protein in model group was not significantly different;Compared with the model group,there was no significant difference between the dosages of Cistanche deserticola and the mosapride group.5.Pathology:Compared with the blank group,the colonic mucosal structure of rats in the model group remained intact without significantly impaired was observed in the submucosal layer of the colon,and inflammatory cells in the muscularis propria were increased.Cistanche deserticola dose groups and mosapride groups had no significant changes compared with blank group.Conclusion: 1.The rat model of functional constipation was established by placing the rat in the freezer in this experiment,and the operation was simple.2.The prevention and treatment of functional constipation caused by low temperature environment by Cistanche deserticola may be related to the promotion of gastrointestinal peristalsis and the improvement of intestinal neurotransmitters.3.The high dose group of Cistanche deserticola had better control effect than other groups,which could be further popularized in clinical practice.
