Background:Eukaryotic translation elongation factor 18(EEF1D),a subunit of the protein elongation factor 1 complex,mediates the elongation process of protein synthesis.In addition to this classical biological function,EEF1 D is upregulated in many tumors,such as liver cancer,oral squamous cell carcinoma and medulloblastoma.EEF1D is involved in the pathogenesis and progression of a variety of tumors.Overexpression of EEF1D can promote the migration and invasion of oral squamous cell carcinoma via activating the EMT signaling pathway.Moreover,it can regulate the proliferation of osteosarcoma through the Akt-mTOR signaling pathway.These results indicate that EEF1D plays a very important role in the development of tumors.However,the effect of EEF1d on the malignant phenotype of glioma and its specific influencing mechanism have not been studied.Methods:The different mRNA expression level of EEF1D between tumor tissues and non-tumor brain tissues was analyzed by using TGCA data.Then,quantitative real-time PCR,immunohistochemistry and Western Blot were used to detect the expression of EEF1D in clinical specimens.CCK8 assay was used to detect cell proliferation in vitro,and Transwell assay and Boyden assay were used to detect cell migration and invasion ability,respectively.In addition,we investigated the effect of EEF1D on EMT and PI3K/Akt pathway in glioma by Western blot.Finally,protein immunoprecipitation combined with mass spectrometry were used to predict the potential proteins cross binding with EEF1D,and GO and KEGG enrichment analysis was used to further predict the function of EEF1D.Results:We found that the expression of EEF1D was up-regulated in glioma tissues,and could promote the proliferation,migration and invasion of glioma.Further studies showed that knockdown of EEF1D expression could decrease the activity of EMT and PI3K/Akt pathway.Finally,the results of enrichment analysis showed that EEF1D was multifunctional and involved in many biological processes of glioma,but mainly concentrated in nucleic acid metabolism and cell growth.Therefore,EEF1D may be involved in the process of DNA replication and RNA synthesis by binding catalytic proteins and regulating their activity.Conclusion:In this study,we identified that EEF 1D could be a new target for glioma and predicted the potential pathway of this target in promoting the development and process of glioma.It was suggested that EEF 1D has the potential to be a new prognostic indicator and therapeutic target of glioma.
