Objective:In this study,we tried to observe the effects of acupuncture combined with five elements music therapy on body weight,behavior and the contents of 5-HT and NE in hypothalamus,prefrontal cortex and hippocampus of rats with post-stroke depression,so as to provide scientific basis for acupuncture combined with five elements music therapy in the treatment of post-stroke depression.Method:Fifty SPF healthy male rats weighing 250-300g were randomly divided into blank group,model group,music group,acupuncture group and acupuncture sound group,with 10 rats in each group.Except the blank group,the other four groups were treated with middle cerebral artery occlusion(MCAO)according to Koizumi’s method.The rats with neurological function score of 1-3 points after MCAO were selected.On the third day after MCAO recovery,chronic mild unpredictable stress(CUMS)combined with solitary culture method was used to prepare PSD model rats.The blank group was fed normally throughout the experiment,without modeling or intervention,the model group was only modeling without treatment,and the treatment group was treated on the 21st day of CUMS;once a day,once a course of treatment for 7 days,and one day off between courses of treatment for a total of 2 courses.The music group was treated with music playing,the acupuncture group was treated with "Baiui",and the acupuncture group was treated with acupuncture on the basis of music therapy.The body weight and behavior of rats in each group were observed after intervention.After the treatment,the hypothalamus,prefrontal cortex and hippocampus tissue samples were taken,and the content of NE and 5-HT in hypothalamus,prefrontal cortex and hippocampus tissue was detected by HPLC-ECD.Results:1.Two hours after MCAO model was established,the neurological deficit score was performed after the rats were awake.Four rats that did not meet the score were excluded,and the longa score of the remaining 36 rats was 1-3.After 24 hours,the neurological deficit of the rats basically returned to normal,and the longa score was 0.It was said that the rats had hemiplegia on the contralateral limb of the lesion after anesthesia.2.Effect of acupuncture sound combined therapy on body weight and behavior of rats with post-stroke depression:compared with the blank group,there were significant differences in body weight and behavior indexes of rats in the model group(P<0.01),suggesting that PSD rat model was successfully established.After the intervention,compared with the model group,there were significant differences in body weight and behavioral indicators between the music group and the acupuncture group(P<0.01),while there were significant differences in the acupuncture group(P<0.05).Compared with the acupuncture group,the acupuncture sound group had significantly lower body weight(P<0.01),higher sucrose preference(P<0.05)and significantly reduced swimming rest time(P<0.05).There was no significant difference between the acupuncture sound group and the music group(P>0.05).3.Effect of acupuncture sound combined therapy on NE and 5-HT contents in hypothalamus,prefrontal cortex and hippocampus of post-stroke depression rats:compared with blank group,NE content in hypothalamus of model group decreased significantly(P<0.05),The content of 5-HT in hypothalamus,5-HT and NE in prefrontal cortex and hippocampus were significantly decreased(P<0.01).After intervention,compared with the model group,the content of 5-HT and NE in hypothalamus,prefrontal cortex and hippocampus were effectively increased in the three groups,especially in hypothalamus and hippocampus(P<0.01).The NE content in prefrontal cortex was not significantly increased in music group after treatment(P>0.05).The acupuncture sound group significantly increased the content of 5-HT in prefrontal cortex(P<0.05),the content of NE in prefrontal cortex and the content of 5-HT and NE in hypothalamus and hippocampus(P<0.01).Compared with music group,the NE content in prefrontal cortex and 5-HT content in hippocampus were significantly increased in acupuncture sound group(P<0.01).Compared with the acupuncture group,the NE content in prefrontal cortex and 5-HT content in hippocampus were significantly increased in the acupuncture sound group(P<0.05 and P<0.01,respectively).Conclusion:Acupuncture,music of five elements and acupuncture combined with music of five elements can have effective effects on the behavior of PSD model rats,which is mainly manifested in increasing the body weight,sugar preference,horizontal and vertical scores of PSD rats in different degrees,and reducing the immobility time of swimming.From the behavioral point of view,it has antidepressant effect.In addition,the three treatment groups play an antidepressant role by increasing the content of 5-HT and NE in hypothalamus,prefrontal cortex and hippocampus.In addition,acupuncture combined with five elements music group is better than single treatment group in many aspects,which may be related to its extensive activation of different brain regions and multi-target stimulation.
