Objective:Adopt a network pharmacology research method,Obtaining the "component-target"network of the Xu’s Shenqi Yizhu Decoction with gastric cancer,and predicting the molecular biological pathway nature of the Xu’s Shenqi Yizhu Decoction in the treatment of gastric cancer,to verify the predicted results by in vitro experiments,so as to provide basis for further study in the later stage.Methods:Using the ADME parameters of pharmacokinetics as the screening criteria,and the compounds and effective targets of 9 traditional Chinese medicines of the Xu’s Shenqi Yizhu Decoction were collected by means of the systematic pharmacology analysis platform(TCMSP).The drug-component-target-disease network was constructed by using Cytoscape software.The disease targets of gastric cancer screened by GeneCards database were searched,and the effective components of Chinese traditional medicine were analyzed by Venny to determine the target relationship of intersection.Target protein interaction(PPI)network was constructed from String database.Gene function GO analysis and KEGG pathway enrichment analysis were carried out by R software,and the possible functional pathways of the Xu’s Shenqi Yizhu Decoction in the treatment of gastric cancer were obtained.The effects of the Xu’s Shenqi Yizhu Decoction on the proliferation,migration and apoptosis of human gastric cancer BGC-823 cells were studied by MTT,scratch and Western blot.Results:A total of 121 drug active compounds,87 drug targets,2621 gastric cancer related targets and 87 intersection targets were obtained by taking the oral utilization(OB)≥30%and quasi-drug property(DL)≥0.18 as the screening criteria.The constructed PPI network involved 87 protein nodes,with key targets including TP53,MAPK8,CASP8,IL6,ESR1,APP,AR,CCND1,EGFR,etc.GO functional analysis was mainly related to steroid hormone receptor activity,protein homologous dimer activity,RNA polymerase Ⅱ transcription factor binding,proximal promoter sequence specific DNA binding and other aspects.There were 20 KEGG pathways,mainly involving pi3k-akt signaling pathway,MAPK signaling pathway,P53 signaling pathway,hypoxia-inducible factor(HIF)-1 signaling pathway,apoptosis,FoxO(Forkhead box O)signaling pathway,and so on.The results of MTT,Scratch test and Western blot indicated that the Xu’s Shenqi Yizhu Decoction could inhibit the proliferation and migration of human gastric cancer BGC-823 cells and induce their apoptosis.The prediction was basically consistent with the experimental results,indicating that the Xu’s Shenqi Yizhu Decoction was valuable in the treatment of gastric cancer.Conclusion:This study preliminary forecast the Xu’s Shenqi Yizhu Decoction may through PI3K-Akt signaling pathway and MAPK signal pathway,P53 signaling pathway,signaling pathways,FoxO HIF-1 signaling pathways play to the role of the treatment of cancer of the stomach,In vitro experiments also proved that the Xu’s Shenqi Yizhu Decoction could inhibit the proliferation and migration of human gastric cancer BGC-823 cells and induce their apoptosis.The results show that of the Xu’s Shenqi Yizhu Decoction plays a role in the treatment of gastric cancer from multiple targets and pathways,and it also shows that some feasibility of the Xu’s Shenqi Yizhu Decoction in the treatment of gastric cancer by means of network pharmacology,so as to provide basis for further study in the later stage.It provides a new thought and method for the future study of TCM compound.
