| The immune system plays key roles in keeping healthy,including defending the invasion of viruses and pathogenic bacteria and fungi.The disturbances of immune system can cause a huge impact on the health.Traditional Chinses medicines(TCMs)have been considered to have abilities in adjusting the immunological functions.The purpose of this dissertation was to find out the bioactive substances that can regulate T cells from TCMs.In-depth investigation of immunomodulatory compounds would be helpful in research on the pharmacology of TCMs and development of new pharmaceuticals.This dissertation focused on the chemical investigation of three TCMs screened with immunomodulatory bioactivity,and was divided into three chapters.In the first chapter,chemical ingredients and their bioactivities of the medicinal plant Dendrobium nobile were investigated,and 27 compounds(1-27)were isolated and identified from the bioactive fraction.The fraction was prepared from ethanol extract on the microporous resin column eluted with low polarity fraction,and exhibited immunomodulatory bioactivity on T cells.Compound 12 and 27 could significantly regulate T cells at a concentration of 10μg/ml and inhibit the lung cancer H1299 cells,and compound 27 showed better effect than 12,indicating that 27 might be a potential immunomodulatory substance in D.nobile.The chemical ingredients of Daphne genkwain was described in the second chapter.The fraction of the moderate polarity fraction of the crude extract presented bioactivity on the T cells.Six compounds were isolated and elucidated from this fraction.In the third chapter,the chemical research of Crataegus pinnatifida was described.In the immunomodulatory bioactivity test,low polarity fraction showed potential immunomodulatory bioactivities on T cells,and seven compounds were obtained. |
