Objective:Human Umbilical Vein endothelial cells(HUVECs)were injured by iohexanol in vitro,and the protective effect of Shexiang Baoxin pill on HUVECs injured by iohexanol in vitro.Methods:1.Human umbilical vein endothelial cells(HUVECs)were treated with DEMEmedium,DEME medium+iodoxanol(50MGI/ml,100MGI/ml,200MGI/ml)for 1h,3h,6h and 12h in vitro.The morphological changes of cells were observed under light microscope.The level of lactate dehydrogenase(LDH)in cell supernatant was determined by ELISA to detect cell injury;MDA level in cell was measured by ELISA to detect cell oxidation.2.Human umbilical vein endothelial cells(HUVEC)were pretreated with DEME medium containing musk xiangxinprotecting pills(high concentration 2.0g/L,medium concentration 1.0g/L,low concentration 0.5g/L)for 6h,and the cell injury model was established as above.The morphological changes of cells were observed under light microscope.The concentrations of lactate dehydrogenase(LDH),nitric oxide(NO),intracellular malonic acid(MDA)and superoxide dismutase(SOD)in supernatant were determined by ELISA.Results:1.Compared with the control group,iodixanol can cause changes in the morphology of HUVECs and a decrease in cell density,and as the concentration of iodixanol increases,the more obvious the changes in cell morphology,the more obvious the decrease in cell density;the control group LDH,MDA The concentration did not change significantly with time(r=0.051,P>0.05;r=0.048,P>0.05);the levels of LDH and MDA in the experimental group were significantly increased(P<0.01).The increase in the concentration of iodixanol had an effect on human umbilical vein endothelium The cell release of LDH and the production of MDA increased significantly(P<0.01);when the same concentration of iodixanol is used,the concentration of LDH and MDA can increase significantly with the increase of the time of action.2.After 6 hours of pretreatment with Shexiang Baoxin Pills,compared with the control group,there was no significant improvement in the morphological changes and cell density changes of human umbilical vein endothelial cells induced by iodixanol;compared with the control group,the experimental group LDH,The concentration of MDA decreased(P<0.001),the concentration of SOD increased(P<0.001),and the concentration of NO increased after the pretreatment of high-concentration Shexiang Baoxin Pills(P<0.01);the control results of the experimental group showed that the high-concentration Shexiang Baoxin Pill pretreatment with lower concentration of Shexiang Baoxin Pills reduces the LDH concentration more significantly(P<0.001).Pretreatment with different concentrations of Shexiang Baoxin Pills has no significant change in MDA concentration(P>0.05).High concentration Shexiang Baoxin Pills Pretreatment with lower concentration of Shexiang Baoxin Pills increased the NO concentration more significantly(P<0.05).The pretreatment with high concentration of Shexiang Baoxin Pills increased the NO concentration more significantly than the pretreatment with medium and low concentrations of Shexiang Baoxin Pills(P<0.01).Conclusion:1.Iodixanol can induce injury of human umbilical vein endothelial cells in a concentration and time-dependent manner.2.Shexiang Baoxin Pills can alleviate the injury of human umbilical vein endothelial cells induced by iodixanol;3.Shexiang Baoxin Pills may reduce iodixanol-induced human umbilical vein endothelial cell damage by improving endothelial function and reducing oxidative stress.
