Chemical Constituents Of Medicinal Plants And Promoting Endometrial Receptivities Screening

This dissertation consists of two parts.In the first part,the chemical constituents antimicrobial and cytotoxic activities of Chaenomeles speciosa and Illicium verum were studied.The screening and verification of traditional medicinal plant components which could promote endometrial receptivity were carried out in the second part.I Study on chemical constituents of C.speciosa and I.verumEight known compounds were isolated and identified from dried fruit of C.speciosa,in which compounds 1,2,3 and 5 showed weak antibacterial activity.Fifteen known compounds were isolated and identified from dried fruit of I.verum,in which compounds 5,9 and 15 showed weak antibacterial activity.II Screening and verification of traditional medicinal plant components that promote endometrial receptivitiesFractions were made of reported medicinal plants used for infertility,in vitro screened with endometrial receptivity model.Six fractions were obtained by evaluation at transcriptional level,and further re-assayed with protein level,resulting in active fractions from 3 medicinal plants including Epimedium brevicomu.The improvement of endometrial receptivity of fraction of E.folium was carried out with embryo implantation dysfunction model.With progynova as the positive group,the intragastric administration group was set as low（50 mg/kg）,medium（100 mg/kg）and high（200 mg/kg）doses.The results suggested that fraction of E.folium could increase the expression of estrogen,progesterone and LIF protein,increase the number of uterine glands,uterine blood vessels,and the thickness of the endometrium,promote pinopode number and development which are biomarks of endometrial receptivity improvement.The content of estrogen and progesterone,the thickness of endometrium,and the development of pinopode in the treatment group with fraction of E.folium were better than those of the positive drugs.The screening model of traditional medicinal plant components that promote endometrial receptivity was established.This aims to provide possible components from natural products and potential basis for treating infertility.
