| Objective: Objective to explore the effect of Pien Tze Huang on the proliferation and apoptosis of RPMI 8226 cell line(human multiple myeloma cell line),and to study the signal transduction pathway of intervention,objective to find a new treatment scheme for multiple myeloma,and to provide a new way to reduce the side effects of chemotherapy.Methods:(1)Methods: RPMI 8226 cell line(human multiple myeloma cell line)was selected as the experimental object.Different doses of(0,250,400,500,800,1000,1600 μg/ml)were given to the RPMI 8226 cells for 48 h and 72 h respectively.The cell viability of RPMI8226 was detected by CCK-8 method,and the proliferation ability of RPMI 8226 cells was analyzed.(2)Real time PCR was used to detect the effects of Pien Tze Huang of the anti apoptotic genes(Bcl-xl and MCL-1)and pro apoptotic genes(BOK,BAK-1 and BAX)in the RPMI 8226 cells.Flow cytometry was used to detect the apoptosis of RPMI 8226 cells after the application of Pien Tze Huang.(3)The effects of Pien Tze Huang on cell cycle regulatory genes(Cyclin A and Cyclin D)of RPMI 8226 were detected by real-time PCR,and flow cytometry was used to detect the effect of Pien Tze Huang on cell cycle of RPMI 8226.(4)Western blot was used to examine the effects of different concentrations of Pien Tze Huang tablet on PI3K-Akt signaling pathway in RPMI 8226 cells,objective to further explore the specific mechanism of Pien Tze Huang.Results:(1)CCK-8 results showed that PTH could inhibit the proliferation of RPMI 8226 cells,and with the increase of PTH concentration,the survival inhibition rate of RPMI 8226 cells was also further increased,which was positively correlated with PTH concentration.(2)With the increasing concentration of PTH,the apoptosis rate of RPMI 8226 cells increased gradually,the expression of Bcl-xl decreased,the expression of BOK,BAK-1 and BAX increased,which was dose-dependent and time-dependent.The change of Mcl-1 was not obvious.(3)Effects of PTH on cell cycle of RPMI 8226 cells: RPMI8226 cells were treated with different concentrations of PTH(100,200,400 800 μg/ml)for 24 h and 48 h,we found that RPMI 8226 cells were arrested in G1 phase,S phase was significantly shortened,G2 phase was prolonged,and the proliferation of RPMI 8226 cells was inhibited.The results of cycle related genes showed that the expression of Cyclin A protein was up-regulated,but the expression level of Cyclin D in RPMI8226 cells did not change significantly.(4)The results of Western blot showed that the expression levels of PI3K-110 and NF-κB were decreased,compared with the control group,but the expression of AKT was not significantly changed.Conclusion:(1)PTH can significantly inhibit the proliferation of RPMI 8226 cells,and has a positive correlation with concentration and time.(2)PTH can promote the apoptosis of RPMI 8226 cells.(3)When PTH promoted the division of RPMI 8226 cells,it was blocked in G1 cycle.(4)PI3K-AKT signaling pathway was the main mechanism of PTH in RPMI 8226 cells. |
