Objective:By detecting the expression of nectin-3 protein and MMP-9 protein in selected pancreatic ductal adenocarcinoma(PDAC)tissue,pancreatic benign tumor tissue,and normal pancreatic tissue adjacent to cancer,the expression of these two proteins in PDAC tissue is analyzed The relationship with the clinicopathological characteristics of the patient provides new theoretical evidence for the prognostic evaluation of the patient,and at the same time provides a new target for chemotherapy for cancer patients.Methods:Retrieving the basic clinical pathological information of patients with pancreat-ic tumors submitted by the Department of Hepatobiliary Surgery from the First Affiliated Hospital of Wannan Medical College/Yijishan Hospital.The specimens are all from the First Affiliated Hospital of Wannan Medical College/Yijishan Hospital in 2016 From January to January 2020,a total of 89 cases were surgically resected and pathologically diagnosed as PDAC.None of the selected patients received adjuvant treatments such as radiotherapy,chemotherapy,immune,and targeted therapy before surgery.In addition,there were 12 cases diagnosed as pancreatic mucinous cystic tumor and serous cystadenoma during this period.The selected cases have complete clinical pathological data.At the same time,10 cases of normal pancreatic tissue adjacent to cancer were randomly selected.Analyze the expression of nectin-3 and MMP-9 in the tissue wax block of selected cases,as well as the relationship between the two proteins and some clinicopathological characteristics of the patient,and analyze the relationship between nectin-3 protein and MMP-9 protein The correlation between PDAC tumor invasion,metastasis and prognosis.Results:1.The results of immunohistochemical staining showed that both nectin-3 protein and MMP-9 protein were expressed in the cytoplasm and/or cell membrane.Through detection,the positive expression rate of nectin-3 protein in the tissues of PDAC cases was 87.64%(78/89),the positive expression rate in pancreatic benign tumors was 25.0%(3/12),and the positive expression rate in normal pancreatic tissues adjacent to cancer was 10.0%(1/10).The positive expression rate of MMP-9 protein in PDAC case tissue was 86.52%(77/89),the positive expression rate in pancreatic benign tumor case tissue was 25.0%(3/12),and the positive expression rate in normal pancreatic tissue adjacent to cancer It is 10.0%(1/10).The results showed that the expression rate of nectin-3 protein in PDAC tissues was significantly higher than that of benign pancreatic tumors and normal pancreatic tissues adjacent to cancer,and the difference was statistically significant(x~2=40.936,P<0.05).The expression rate of MMP-9 protein in PDAC tissues was significantly higher than that of benign pancreatic tumors and normal pancreas adjacent to cancer tissues,and the difference was statistically significant(x~2=39.153,P<0.05).2.The expression of Nectin-3 protein in PDAC tumor tissues has a certain correlation with p TNM staging,lymph node metastasis,vascular invasion and nerve invasion of PDAC tumors,and the difference is statistically significant(P<0.05);Nectin-3 protein in PDAC tumors There was no significant correlation between the expression in the tissue and some clinicopathological characteristics of patients,such as age,gender,tumor location,tumor size and tumor differentiation,and the difference was not statistically significant(P>0.05).The expression of MMP-9 protein in PDAC tumor tissue has a certain correlation with the p TNM stage,lymph node metastasis and vascular invasion of PDAC tumor,and the difference is statistically significant(P<0.05).There is no significant correlation between the expression of MMP-9 protein in PDAC tumor tissue and the patient’s clinicopathological characteristics,such as age,gender,tumor location,tumor size,tumor differentiation,and whether there is neuroinvasion,and there is no statistical difference.Academic significance(P>0.05).3.There is no correlation between the expression of nectin-3 and MMP-9 in pancreatic ductal adenocarcinoma(PDAC)tissues(r=1.0,P>0.05).Conclusion:1.The expression rate of Nectin-3 protein and MMP-9 protein in normal pancreatic tissue adjacent to cancer,pancreatic benign tumor tissue and PDAC tissue gradually increased,indicating that Nectin-3 protein and MMP-9 protein are involved in the occurrence and development of pancreatic tumors Play a role in promoting;2.Among PDAC cases,p TNM staging is stage III and IV,with regional lymph node metastasis,vascular invasion and nerve invasion,the expression rate of nectin-3 protein is higher;indicating the prognosis of nectin-3 protein and PDAC closely related;3.Among the PDAC cases,the p TNM stage is III and IV,and the PDAC cases with regional lymph node metastasis and vascular invasion have a higher expression rate of MMP-9 protein;indicating that MMP-9 protein is closely related to the prognosis of PDAC.4.The expression levels of Nectin-3 protein and MMP-9 protein in PDAC tissues do not show a significant correlation,suggesting that the two may not have a synergistic effect in the occurrence and development of PDAC.
