Objective:By examining the expression levels of AGEs,RAGE,IL-6 and CXCL12 in the uterosacral ligament tissue of patients with and without the pelvic organ prolapse,and comparing whether there are differences and correlations between them,to explore the pathogenesis of the disease and provide a new idea for the prevention and treatment of it.Methods:A total of 32 cases of fresh sacral ligament tissue from patients undergoing hysterectomy for POP and 30 cases from patients undergoing hysterectomy for other benign gynecological diseases were collected from gynecological inpatients in Yijishan Hospital of Wannan Medical University,from September 2019 to September 2020,and theyweredividedintoPOPgroupandCON group,respectively.Immunohistochemistry(Envision two-step method)was used for localization and quantitative analysis of AGEs and RAGE.Western Blot was used to detect the relative protein content of RAGE in the two groups.ELISA was used to detect the concentration levels of AGEs,IL-6 and CXCL12 in the two groups.Software such as Image J,Excel,SPSS 26.0 and Graph Pad Prism 7.0 were used to collect data,analyze differences and correlations,and make statistical charts.Results:A total of 32 participants were enrolled in the POP group and 30 participants in the CON group.Immunohistochemical results showed that protein expression levels of RAGE and AGEs in POP group were higher than those in CON group,with statistical significance(p<0.05).The results of Western Blot showed that the expression of RAGE/β-actin in the POP group was 0.945±0.195,and that in the CON group was0.504±0.060.The relative expression level of RAGE in the POP group was higher than that in the CON group,and the difference was statistically significant(p<0.05).ELISA results showed that compared with the CON group,the concentrations of AGEs,IL-6 and CXCL12 were higher in the POP group,and the difference was statistically significant(p<0.05).Pearson correlation analysis showed that AGEs was positively correlated with the expression of IL-6 and CXCL12,and the correlation was significant(r IL-6=0.857,r CXCL12=0.778,p<0.01).Conclusion:1.POP occurs with AGEs and RAGE.2.The expression of inflammatory factors(IL-6,CXCL12)was increased in POP,which suggests that inflammatory cells were infiltrated in POP.3.There was a significant correlation between the expression of AGEs and inflammatory factors(IL-6,CXCL12)in POP,suggesting that AGEs/RAGE may induce the occurrence of POP by increasing inflammatory cell infiltration.Anti-inflammatory molecular targeting inhibition of AGEs may be one of the ideal strategies for the prevention and treatment of POP.
