Objective: To observe the effects of Qutan Huoxue Decocotion on the expression of micro RNA-27a(miR-27a),p38 mitogen-activated protein kinase(p38MAPK)pathway and aquaporin-9(AQP9)in the liver tissues of rats with non-alcoholic fatty liver disease(NAFLD).Methods: Choosing 32 clean grade male SD rats,weight in 180~220g with normal diet adaptive feeding one week.All rats were randomly divided into two groups:normal group(n=6),and model group(n=26).indecing NAFLD model of rats was by feeding a high-fat diet.8 weeks later,two rats from the model group were randomly killed,whether the mold is successful or not.The remaining molds(24)were randomly divided into 4 groups,each group of six,then dividing the remaining molds into model group,Qutan Huoxue Decocotion low dose group,middle dose group and high dose group(5.4g/kg,10.8g/kg,21.6g/kg)randomly.They were executed after 4 weeks of continuous intervention.automatic biochemical analyzer detected Serum levels of ALT,AST,TG,TC,LDL and HDL;observing the pathological changes of liver used HE staining and oil red O staining.The mRNA expression levels of miR-27 a,AQP9 and p38MAPK in liver tissues of each group were detected by real-time PCR;The protein expression levels of AQP9,p38MAPK and p-p38MAPK in liver tissues of each group were detected by Western-blot.Results: 1.The results of serum biochemical indexes:Compared with normal group,serum ALT,AST,TG,TC,LDL indexes in the model group were evidently ascend,while HDL descend(P<0.05);Compared with model group,serum AST,ALT,TG,TC,LDL levels in Qutan Huoxue Decocotion groups were significantly descend,while HDL ascend(P<0.05),the effect was most pronounced in the high-dose group.2.HE staining and oil red O staining showed that,compared with the normal group,model group had more steatosis and more fat droplets in liver cells of rats;Compared with model group,Qutan Huoxue Decocotion group had less steatosis and less fat droplets,the effect was most pronounced in the high-dose group.3.Realtime-PCR results showed that,compared with the normal group,the model group showed the expression of miR-27 a were higher in liver tissue,and the mRNA expressions of AQP9 and p38MAPK were significantly increased(P<0.05).Compared with model group,The expression of miR-27 a in liver tissues increased,while the expression of AQP9 and p38MAPK mRNA decreased(P<0.05),Compared with Low dose group and middle dose group,the effect was most pronounced in the high-dose group.4.Protein expression: Compared with normal group,the protein of rats in model group had higher expression levels of AQP9 and p-p38MAPK in liver tissue;Compared with model group,the protein of rats in Qutan Huoxue Decocotion group had lower expression levels of AQP9 and p-p38MAPK in liver tissue(P<0.05).Conclusion: 1.Qutan Huoxue Decocotion can reduce serum ALT,AST,TG,TC,LDL levels in NAFLD,elevate HDL levels;2.Qutan Huoxue Decocotion can alleviate liver steatosis and reduce the number of lipid droplets in NAFLD rats;3.Qutan Huoxue Decocotion up-regulated the expression of miR-27 a and down-regulated the expression of p38MAPK and AQP9 mRNA;4.Qutan Huoxue Decocotion can down-regulate the expression of p-p38MAPK,and then reduce the expression level of AQP9,and finally reduce the formation and accumulation of liver triglyceride,cholesterol and other lipid substances,and finally reduce the degree of liver steatosis.
