ObjectiveMalignancies affecting the breast are prevalent among the female population and a relatively common cause of death.GRB-2 related adaptor protein 2(GRAP2)is a leukocyte-specific adaptor protein which acts as a central connexin in the activation of immune cells.Recent evidence have reported that GRAP2 suppresses the growth of tumor cells despite its low expression in these cells.This study aims to explore the expression of GRAP2 in breast cancer and verify the correlation in survival and prognosis of breast cancer patients by statistical analysis and in vitro experiments.MethodsPathological GRAP2 expression in breast tumors was examined by exploring TCGA-BRCA level 3 data.ROC curve was drown to evaluate the diagnostic efficacy of GRAP2 in breast cancer.The chi-squared test was set to evaluate the relationship between clinicopathological data and GRAP2 expression.GRAP2 expression effects were examined with survival and Cox regression models to appraise their impact on breast patient’ overall survival and relapse free survival rate.The GSE project in bc-Gen Ex Miner v4.2 was used to verify the findings.Western blotting test and Real-Time quantitative polymerase chain reaction(RT-q PCR)were used to verify the expression of GRAP2 in breast cancer.ResultsThe clinical data from 1104 cases of primary breast tumors showed that they had low GRAP2 expression,and GRAP2 expression level has certain diagnostic value in breast cancer.The expression of GRAP2 has certain statistical significance with the clinicopathological characteristics of breast cancer patients.Univariate and multivariate cox analysis showed that GRAP2 can directly impacted breast cancer prognosis.GRAP2 OS and RFS of patients with low expression were significantly shortened.Western Blot and RT-q PCR results showed that the mRNA and protein expression in cancer cells were lower than that in normal mammary gland.ConclusionGRAP2 expression is reduced in breast cancer,and low GRAP2 breast tumor expression is a novel,independent prognosticator of poor patient outcomes.
